本文通过荧光滴定、紫外可见吸收滴定、紫外熔点实验以及CD光谱研究了三联吡啶衍生物L1-L3及其Pt(Ⅱ)配合物1-3和人端粒i-motif DNA的相互作用。结果表明:配体和配合物均能与i-motif DNA相互作用,结合常数约为105 M-1,而且配合物的亲和作用强于配体。配合物的结合导致i-motif DNA的熔点温度降低并对其构象产生扰动。
本文通過熒光滴定、紫外可見吸收滴定、紫外鎔點實驗以及CD光譜研究瞭三聯吡啶衍生物L1-L3及其Pt(Ⅱ)配閤物1-3和人耑粒i-motif DNA的相互作用。結果錶明:配體和配閤物均能與i-motif DNA相互作用,結閤常數約為105 M-1,而且配閤物的親和作用彊于配體。配閤物的結閤導緻i-motif DNA的鎔點溫度降低併對其構象產生擾動。
본문통과형광적정、자외가견흡수적정、자외용점실험이급CD광보연구료삼련필정연생물L1-L3급기Pt(Ⅱ)배합물1-3화인단립i-motif DNA적상호작용。결과표명:배체화배합물균능여i-motif DNA상호작용,결합상수약위105 M-1,이차배합물적친화작용강우배체。배합물적결합도치i-motif DNA적용점온도강저병대기구상산생우동。
The interactions of terpyridine derivatives(L1-L3)and their Pt(Ⅱ)complexes(1-3)with human telomeric i-motif DNA were studied by UV-visible absorption titration,fluorescence titration,UV melting,and CD spectroscopy. The results show that these compounds can bind to i-motif DNA,and the affinity potential of complexes to i-motif DNA is larger than ligands,with the binding constants(Kb)of 3. 20(3. 80(105and 1. 20(1. 43(105M-1,respectively. UV-melting and CD spectral results indicate that the bind-ing of complex destabilizes the structure of i-motif DNA,while the binding of ligands fails to change the conformation and the melt-ing temperature of DNA.
