目的:观察不同剂量水蛭活血散肿口服液对大鼠脑缺血再灌注损伤的改善作用。方法将SD大鼠随机分为6组,分别为假手术组(手术不结扎血管,不注射硝普钠)、模型对照组、水蛭活血散肿口服液低剂量组(2 g生药/kg)、水蛭活血散肿口服液中剂量组(4 g生药/kg)水蛭活血散肿口服液高剂量组(8 g生药/kg)及通心络胶囊组(1 g/kg)。各给药组均灌胃给药,容量为1 mL/100 g体质量。假手术组及模型对照组灌胃等容积蒸馏水,连续7 d。在末次灌胃后2 h,在10%水合氯醛麻醉下(350 mg/kg,腹腔注射),分离并用无损伤动脉夹夹闭大鼠两侧颈动脉,同时迅速腹腔注射硝普钠(2.5 mg/kg,腹腔注射),缺血45 min后,将颈总动脉松开,再灌注30 min。实验结束腹主动脉取血2 mL,3000 r/min,4℃离心10 min,取血清按说明书方法测定其中超氧化物歧化酶( SOD)、丙二醛( MDA)、一氧化氮( NO)含量。结果水蛭活血散肿口服液(2 g、4 g、8 g生药/kg)可升高脑缺血再灌注损伤大鼠血清中NO、SOD含量,降低MDA含量,纠正氧自由基代谢紊乱。结论水蛭活血散肿口服液对大鼠脑缺血再灌注损伤具明显的改善作用。

목적:관찰불동제량수질활혈산종구복액대대서뇌결혈재관주손상적개선작용。방법장SD대서수궤분위6조,분별위가수술조(수술불결찰혈관,불주사초보납)、모형대조조、수질활혈산종구복액저제량조(2 g생약/kg)、수질활혈산종구복액중제량조(4 g생약/kg)수질활혈산종구복액고제량조(8 g생약/kg)급통심락효낭조(1 g/kg)。각급약조균관위급약,용량위1 mL/100 g체질량。가수술조급모형대조조관위등용적증류수,련속7 d。재말차관위후2 h,재10%수합록철마취하(350 mg/kg,복강주사),분리병용무손상동맥협협폐대서량측경동맥,동시신속복강주사초보납(2.5 mg/kg,복강주사),결혈45 min후,장경총동맥송개,재관주30 min。실험결속복주동맥취혈2 mL,3000 r/min,4℃리심10 min,취혈청안설명서방법측정기중초양화물기화매( SOD)、병이철( MDA)、일양화담( NO)함량。결과수질활혈산종구복액(2 g、4 g、8 g생약/kg)가승고뇌결혈재관주손상대서혈청중NO、SOD함량,강저MDA함량,규정양자유기대사문란。결론수질활혈산종구복액대대서뇌결혈재관주손상구명현적개선작용。
Objective To study the effect of different dose of Leeches Huoxue Sanzhong oral liquid on cere -bral ischemia-reperfusion injury in rats.Methods SD rats were randomly divided into six groups ,sham operation group( operation without vascular ligation and injection of sodium nitroprusside ) ,cerebral ischemia-reperfusion mod-el control group,Leeches Huoxue Sanzhong oral liquid low dose group (2 g crude drug/kg),Leeches Huoxue Sanzhong oral liquid middle dose group(4 g crude drug/kg),Leeches Huoxue Sanzhong oral liquid high dose group (8 g crude drug/kg) and Tongxinluo capsule group (1 g/kg).Each treatment group was administered orally with a capacity of 1 mL/100 g body mass.Sham group and model control group was given the same volume of distilled water for 7 d.Af-ter the last administration at 2 h,cerebral ischemia-reperfusion injury model was established .End of the experiment abdominal aortic blood was collected for determination of SOD ,MDA and NO.Results Leeches Huoxue Sanzhong o-ral liquid(2 g,4 g and 8 g crude drug/kg) can increase contents of NO and SOD in serum of rat with cerebral ische-mia-reperfusion injury ,decrease MDA level ,and correct oxygen free radicals metabolic disorders .Conclusion Lee-ches Huoxue Sanzhong oral liquid has obvious improving effect on ischemia reperfusion injury in rat brain .
