目的:观察鼻咽癌组织中hMLH1基因启动子区甲基化状态及其对蛋白表达的影响,并分析甲基化及蛋白表达与临床病理特征的关系。方法采用甲基化特异性定量PCR检测鼻咽癌组织及鼻咽慢性炎症组织中hMLH1的甲基化状态。应用免疫组化法检测hMLH1蛋白表达情况。结果鼻咽癌组织和鼻咽慢性炎症组织中 hMLH1基因甲基化阳性率分别为77.8%(57/63)、8.3%(1/12)(P<0.001)。hMLH1蛋白表达下调/缺失率分别为71.4%(45/63)、8.3%(1/12)(P<0.001)。hMLH1基因甲基化阳性率与鼻咽癌的临床分期、组织分化程度、肿瘤直径、淋巴结转移无明显相关性(P>0.05),hMLH1蛋白表达与鼻咽癌的淋巴结转移具有相关性(P<0.05)。结论 hMLH1甲基化及其蛋白表达与鼻咽癌关系密切,并且甲基化影响其蛋白表达。

목적:관찰비인암조직중hMLH1기인계동자구갑기화상태급기대단백표체적영향,병분석갑기화급단백표체여림상병리특정적관계。방법채용갑기화특이성정량PCR검측비인암조직급비인만성염증조직중hMLH1적갑기화상태。응용면역조화법검측hMLH1단백표체정황。결과비인암조직화비인만성염증조직중 hMLH1기인갑기화양성솔분별위77.8%(57/63)、8.3%(1/12)(P<0.001)。hMLH1단백표체하조/결실솔분별위71.4%(45/63)、8.3%(1/12)(P<0.001)。hMLH1기인갑기화양성솔여비인암적림상분기、조직분화정도、종류직경、림파결전이무명현상관성(P>0.05),hMLH1단백표체여비인암적림파결전이구유상관성(P<0.05)。결론 hMLH1갑기화급기단백표체여비인암관계밀절,병차갑기화영향기단백표체。
Objective To observe the aberrant methylation and protein expression of hMLH1 gene in nasopharyngeal carcinoma and analyse the relationship between the methylation and clinical manifestation. Methods Methylation-specific PCR (MSP) and the immunohistochemical method were employed to detect promoter methylation and protein expression of the hMLH1 gene in nasopharyngeal cancer tissues and their adjacent tissues from 63 patients. Results Promoter methylation of hMLH1 were found in 77.8%(57/63) and 8.3%(1/12) of the nasopharyngeal carcinoma tissues and chronic inflammation nasopharyngeal tissue (P<0.001). hMLH1 protein expression cut/loss rate was 71.4%(45/63), 8.3%(1/12) (P<0.001). The positive rate of hMLH1 methylation was nonsignificant related to clinical stage, tissue differentiation, tumor size, lymphonode metastasis of carcinoma(P>0.05). hMLH1 protein expression was significant related to lymphonode metastasis(P<0.05). Conclusions Aberrant methylation of the promoter of hMLH1 gene are a common and specifically even in occurrence and progression of nasopharyngeal cancer.Loss of hMLH1 protein is associated with methylation of its promoter.
