采用超声波和微波技术提取大火草[Anemone tomentosa(Maxim.)Péi]根、茎、叶中的多糖,确定超声波醇提为最佳提取方法,用苯酚-硫酸法测定其多糖含量。通过单因素试验和响应面分析法,研究了料液比、温度、提取时间对多糖提取率的影响。结果表明,提取时间和温度是影响大火草多糖提取率的主要因素,试验稳定性、精密度良好,提取大火草多糖的最佳条件为温度80℃,提取时间50 min,料液比1:100(m:V)。利用最佳提取工艺得到根、茎、叶中的多糖含量分别为5.16%、1.61%、7.16%。

The polysaccharides were extracted from the roots, stems and leaves of Anemone tomentosa (Maxim.) Péi with ul-trasonic and microwave extraction. The ultrasound-assisted extraction with ethanol was the best method. Polysaccharides were measured by phenol-sulfuric acid method. The effects of material-liquid ratio, extraction temperature and time on the extrac-tion of polysaccharides were studied with single-factor experiment and response surface methodology. The results showed that the temperature and the time were the main factors. The optimal condition of polysaccharides extraction was temperature of 80℃,extraction time of 50 min, material-liquid ratio of 1:100 (m:V). Under the optimal conditions, the contents of polysaccha-rides in roots, stems and the leaves were 5.16%,1.61% and 7.16%,respectively.
