目的:检测蛛网膜下腔出血(以下简称 SAH)后迟发性脑血管痉挛中TNF-α和NF-кB的表达变化。方法56只中国白兔随机分成3组:对照组( n =8):不做枕大池穿刺注血,处死;SAH组( n =24):在第0天和第2天分二次枕大池注射自体动脉血,分别在第1次注血后第4天、第7天和第14天分三批处死,每组8只;假手术组( n=24):只做枕大池穿刺不注血,第4天、第7天和第14天处死,每组8只。通过计算动脉口径比判断基底动脉痉挛程度,使用免疫组化和免疫蛋白印迹的方法检测迟发性脑血管痉挛中TNF-α和NF-кB的表达变化。结果枕大池注血方法可以造成SAH后迟发性脑血管痉挛模型。 NF-κB 和 TNF-α在注血后第4天表达开始增加( P <0吵.01),在第7天达到最高值( P <0.01),第14天降到对照组水平。结论蛛网膜下腔出血后迟发性脑血管痉挛中NF-κB及 TNF-α表达增加,炎症反应在此过程中起着重要作用。

목적:검측주망막하강출혈(이하간칭 SAH)후지발성뇌혈관경련중TNF-α화NF-кB적표체변화。방법56지중국백토수궤분성3조:대조조( n =8):불주침대지천자주혈,처사;SAH조( n =24):재제0천화제2천분이차침대지주사자체동맥혈,분별재제1차주혈후제4천、제7천화제14천분삼비처사,매조8지;가수술조( n=24):지주침대지천자불주혈,제4천、제7천화제14천처사,매조8지。통과계산동맥구경비판단기저동맥경련정도,사용면역조화화면역단백인적적방법검측지발성뇌혈관경련중TNF-α화NF-кB적표체변화。결과침대지주혈방법가이조성SAH후지발성뇌혈관경련모형。 NF-κB 화 TNF-α재주혈후제4천표체개시증가( P <0묘.01),재제7천체도최고치( P <0.01),제14천강도대조조수평。결론주망막하강출혈후지발성뇌혈관경련중NF-κB급 TNF-α표체증가,염증반응재차과정중기착중요작용。
Objective To observe the expression of TNF-a and NF-кB in delayed cerebral vasospasm ( DCVS) after subarachnoid hemorrhage ( SAH).Methods Fifty-six Chinese rabbits were randomly divided into three groups :① control group ( n =8),the rabbits were not injected;②SAH group ( n =24),autologous arterial blood was injected into the cisterna magna at day 0 and day 2,and eight animals were killed respectively at day 4,day 7,and day 14.③Sham-operation group ( n=24),the rabbits were punctured,but not injected,and eight animals were killed respectively at day 4,day 7,and day 14.Vasospasm degree was evaluated by calculating diameter and thickness of vertebrobasilar arteries .The expression levels of TNF-αand NF-кB in delayed cerebral vasospasm were detected by immunohistochemistry and Western Blot .Results The animal models with delayed cerebral vasospasm after SAH could be established by blood perfusion into cisterna magna .The expression levels of NF-κB and TNF-αwere increased from day 4 ( P <0.01),which reached peak at day 7 ( P <0.01), and decreased to the levels of control group at day 14.Conclusion The expression levels of TNF-αand NF-кB are increased in delayed cerebral vasospasm after subarachnoid hemorrhage , and inflammatory reaction plays an important role during the process.
