目的:制作亚急性肝内胆汁淤积动物模型,探索亚急性胆汁淤积发生的生化和病理变化。方法幼年 SD 大鼠,分为对照组与 ANIT 模型组,分别给与芝麻油以及α-萘异硫氰酸酯(ANIT)灌胃,每周一次共4次,48小时后分批处死动物,取血标本送检肝功能指标,留取肝脏标本观察肝脏以及胆道病理改变。结果ANIT 灌胃后48小时,血转氨酶、胆红素明显上升,肝脏出现结构坏死,炎症细胞浸润、增生,胆管增生,9天、16天逐渐加重,23天肝功能指标逐渐恢复,肝脏结构好转,胆管增生和纤维化仍明显。结论成功制作幼鼠亚急性胆汁淤积的动物模型, ANIT 每周一次灌胃后2~3周内胆汁淤积明显,第4周生化和病理结构逐渐恢复,但胆管增生和肝纤维化仍然明显。

목적:제작아급성간내담즙어적동물모형,탐색아급성담즙어적발생적생화화병리변화。방법유년 SD 대서,분위대조조여 ANIT 모형조,분별급여지마유이급α-내이류청산지(ANIT)관위,매주일차공4차,48소시후분비처사동물,취혈표본송검간공능지표,류취간장표본관찰간장이급담도병리개변。결과ANIT 관위후48소시,혈전안매、담홍소명현상승,간장출현결구배사,염증세포침윤、증생,담관증생,9천、16천축점가중,23천간공능지표축점회복,간장결구호전,담관증생화섬유화잉명현。결론성공제작유서아급성담즙어적적동물모형, ANIT 매주일차관위후2~3주내담즙어적명현,제4주생화화병리결구축점회복,단담관증생화간섬유화잉연명현。
Objective To create the sub-acute intrahepatic cholestasis model with infantile rats and explore the bio-chemical and pathological changes of sub-acute intrahepatic cholestasis. Methods Male SD infantile rats (1 50 ~1 80 g)were randomly divided into two groups.The ANIT (alpha-naphthyl-isothiocyanate)model group was gavaged with four weekly doses of ANIT (80mg/kg),the control group was gavaged with placebo sesame oil.Eight rats from the cholestasis group were sacrificed after 48h of the ANIT treatment every week.The blood samples were collected to assess AST,ALT,TBill,DBill,TBA,ALP and γGT,and the liver tissue samples were made into paraffin sections and observed with microscope. Results The level of serum ALT,TBill,DBill,TBA,ALP and γGT was increased 48h after the ANIT treatment,and maintained at the high level un-til the third week.In the fourth week,the serum ALT,TBill,DBill and ALP declined quickly,however the serum TBA and γGT remained rising.Pathological changes were obvious 48h after the first ANIT treatment,characterizing with liver necrosis,inflam-matory infiltration,bile duct hyperplasia,which appeared gradually increasing until the third week.In the fourth week,the liver necrosis and inflammatory infiltration recovered normally,but the bile duct hyperplasia and liver fibrosis remained aggravating. Conclusion The sub-acute intrahepatic cholestasis model was created successfully with the ANIT gavaging weekly.The chole-stasis changes were the most obvious in the second and third week,but recovered in the fourth week except the bile duct hyper-plasia and liver fibrosis.
