应用本实验室筛选得到的纤维素酶高产菌株里氏木霉( Trichoderma reesei)150-1-1发酵制备纤维素酶固体曲,通过优化固态发酵培养基组成及发酵条件,制备得到纤维素酶活力较高的固体曲及粗酶液。实验结果表明,在原料量为10 g,麸皮与秸杆粉质量比为4:1,料液比为1:2.5,发酵时间为120 h,发酵温度为31℃,发酵起始pH值为5.5的条件下,应用里氏木霉150-1-1发酵得到的纤维素酶固体曲酶活力达到423.6 U/g。
應用本實驗室篩選得到的纖維素酶高產菌株裏氏木黴( Trichoderma reesei)150-1-1髮酵製備纖維素酶固體麯,通過優化固態髮酵培養基組成及髮酵條件,製備得到纖維素酶活力較高的固體麯及粗酶液。實驗結果錶明,在原料量為10 g,麩皮與秸桿粉質量比為4:1,料液比為1:2.5,髮酵時間為120 h,髮酵溫度為31℃,髮酵起始pH值為5.5的條件下,應用裏氏木黴150-1-1髮酵得到的纖維素酶固體麯酶活力達到423.6 U/g。
응용본실험실사선득도적섬유소매고산균주리씨목매( Trichoderma reesei)150-1-1발효제비섬유소매고체곡,통과우화고태발효배양기조성급발효조건,제비득도섬유소매활력교고적고체곡급조매액。실험결과표명,재원료량위10 g,부피여갈간분질량비위4:1,료액비위1:2.5,발효시간위120 h,발효온도위31℃,발효기시pH치위5.5적조건하,응용리씨목매150-1-1발효득도적섬유소매고체곡매활력체도423.6 U/g。
The cellulase solid koji was fermented by using high yielding strain Trichoderma reesei 150-1-1 screened in our laboratory. Solid koji and crude enzyme with high cellulase activities were obtained by optimizing the medium components and fermentation conditions of solid fermentation. The results showed that the cellulase activities of solid koji reached 423. 6 U/g under the optimal conditions where the fermentation material was 10 g,the ratio of straw powder to wheat bran was 4:1,the ratio of materials to water was 1:2. 5,the fermentation reaction temperature was 31℃,the reaction time was 120 h,and the original pH was 5. 5.
