目的:研究中药单体蛇床子素( Osthole, Ost)与5-FU ( Fluorouracil,5-FU)在人胃癌细胞株BGC-823以及人大肠癌细胞株LOVO中的体外相互作用。方法采用MTT法检测单药及联合用药对两株肿瘤细胞株的作用;利用中效原理评价两药之间的相互作用;利用流式细胞仪检测单药及联合用药对肿瘤细胞凋亡和周期的影响。结果Ost和5-FU对两株肿瘤细胞株均具有增殖抑制作用;作用于BGC-823细胞时,当Fa<0.65时, CI<1,两药合用效应为协同作用;作用于LOVO细胞时,当0.05<Fa<0.90时, CI<1,两药合用效应为协同作用。结论 Ost联合5-FU作用于BGC-823细胞在低剂量合用时产生协同效应,协同机制可能与诱导细胞凋亡及G2/M期阻滞相关。

Objective To study the in vitro interactions between osthole ( ost) and fluorouracil ( 5-FU) on human gastric carcinoma cell lines BGC-823 and the human colon cancer cell lines LOVO. Method For single agent and com-bined medication, the effects on two tumor cell lines, the interactions, and the influences on tumor cell cycle and apopto-sis were analyzed using MTT method, median-effect principle and flow cytometry, respectively. Result The combination of ost and 5-FU inhibited the proliferation of the two tumor cell lines; For BGC-823 cells, when Fa<0. 65, CI<1, the combined effects of the two medicine were synergies;For LOVO cells, when 0. 05<Fa<0. 90, CI<1, the effects were also synergies. Conclusion The combined effects of ost and 5-FU on BGC-823 cells were synergistic when in low doses, and the mechanism may be related to apoptosis induced and the block of G2/M phase.
