Longmu Guizhi decoction%ADHD model rats%open-field test%Làt maze
目的:观察龙牡桂枝汤对注意缺陷多动障碍(ADHD)模型动物---幼年自发性高血压大鼠(SHR)的自发活动和非选择性注意力的影响。方法采用幼年雄性SHR大鼠48只,随机分为龙牡桂枝汤高、中剂量组、盐酸哌甲酯组和生理盐水组,连续灌胃给药14 d。通过开场实验和Làt迷宫实验分别检测给药前后大鼠自发活动和非选择性注意力的变化。结果在开场实验中,各药物组穿越格子数和直立次数明显少于生理盐水组,差异有统计学意义(P<0.01);盐酸哌甲酯组大鼠穿越格子数和直立次数少于中药中剂量组,差异有统计学意义(P<0.05),而中药高剂量组与盐酸哌甲酯组比较差异无统计学意义(P>0.05)。Làt迷宫实验中,各药物组斜搭次数多于生理盐水组,差异有统计学意义(P<0.01),中药高剂量组的斜搭次数多于中剂量组及盐酸哌甲酯组,差异有统计学意义(P<0.05)。结论龙牡桂枝汤可改善AD HD模型大鼠的自发活动和非选择性注意力,并呈现出一定的量效关系。

목적:관찰룡모계지탕대주의결함다동장애(ADHD)모형동물---유년자발성고혈압대서(SHR)적자발활동화비선택성주의력적영향。방법채용유년웅성SHR대서48지,수궤분위룡모계지탕고、중제량조、염산고갑지조화생리염수조,련속관위급약14 d。통과개장실험화Làt미궁실험분별검측급약전후대서자발활동화비선택성주의력적변화。결과재개장실험중,각약물조천월격자수화직립차수명현소우생리염수조,차이유통계학의의(P<0.01);염산고갑지조대서천월격자수화직립차수소우중약중제량조,차이유통계학의의(P<0.05),이중약고제량조여염산고갑지조비교차이무통계학의의(P>0.05)。Làt미궁실험중,각약물조사탑차수다우생리염수조,차이유통계학의의(P<0.01),중약고제량조적사탑차수다우중제량조급염산고갑지조,차이유통계학의의(P<0.05)。결론룡모계지탕가개선AD HD모형대서적자발활동화비선택성주의력,병정현출일정적량효관계。
Objective To investigate the effect of Longmu Guizhi decoction (LD) on locomotor activity and non-selective attention of juvenile SHR(spontaneously hypertensive rat)---a rat model of attention def-icit hyperactivity disorder .Methods Forty-eight SHR male were randomly divided into high-dose LD group , middle-dose LD group ,methylphenidate control group and saline control group .All the SHRs were lavaged for 14 days .Locomotor activity and non-selective attention were examined by open-field test and Làt maze test respectively before and after intervention .Results In open-field test ,the number of square crossing and rearing after intervention of methylphenidate and decoctions in SHRs was significantly fewer than the saline group (P<0 .01) .The number of the above items in methylphenidate group was significantly fewer than middle-dose LD group (P<0 .05) .There was no difference between methylphenidate group and high-dose LD group(P>0 .05) .In Làt maze test ,frequency of leaning against the wall after intervention of methyl-phenidate and decoctions in SHRs was significantly increased compared with the saline group (P<0 .01) , and that of high-dose LD group was significantly higher than the methylphenidate group and middle-dose LD group(P<0 .05) .Conclusion Longmu Guizhi decoction can reduce locomotor activity and improve non-se-lective attention of ADHD model rats ,which presents some kind of dose-effect relationship .
