目的:研究丹参多酚酸盐对糖尿病大鼠心肌的保护作用,并探讨可能的作用机制。方法将40只雄性 SD 大鼠随机分为模型组(30只)和空白对照组(10只)。模型组用腹腔注射链脲佐菌素(STZ)的方法建模,将建模成功的大鼠随机分为高剂量丹参多酚酸盐组(10只)、低剂量丹参多酚酸盐组(10只)及对照组(9只),各组均采用腹腔注射给药,持续8周,实验10周后麻醉处死,心脏超声测定大鼠的心脏形态和功能指标,HE 染色观察心肌的病理形态学变化,免疫组化法观察心肌中转化生长因子(TGF -β1)的表达情况。结果与空白对照组相比,模型组大鼠心脏超声各指标明显降低,心肌纤维紊乱,心肌组织中 TGF -β1表达增加。与模型对照组相比,丹参多酚酸盐用药组心脏超声指标及心脏病理情况明显改善,心肌组织中 TGF -β1的表达降低;高剂量组较低剂量组作用更加明显,差异有统计学意义(P ﹤0.05)。结论丹参多酚酸盐对糖尿病大鼠具有明显的心肌保护作用,作用机制可能与降低心肌中 TGF -β1的表达有关。

목적:연구단삼다분산염대당뇨병대서심기적보호작용,병탐토가능적작용궤제。방법장40지웅성 SD 대서수궤분위모형조(30지)화공백대조조(10지)。모형조용복강주사련뇨좌균소(STZ)적방법건모,장건모성공적대서수궤분위고제량단삼다분산염조(10지)、저제량단삼다분산염조(10지)급대조조(9지),각조균채용복강주사급약,지속8주,실험10주후마취처사,심장초성측정대서적심장형태화공능지표,HE 염색관찰심기적병리형태학변화,면역조화법관찰심기중전화생장인자(TGF -β1)적표체정황。결과여공백대조조상비,모형조대서심장초성각지표명현강저,심기섬유문란,심기조직중 TGF -β1표체증가。여모형대조조상비,단삼다분산염용약조심장초성지표급심장병리정황명현개선,심기조직중 TGF -β1적표체강저;고제량조교저제량조작용경가명현,차이유통계학의의(P ﹤0.05)。결론단삼다분산염대당뇨병대서구유명현적심기보호작용,작용궤제가능여강저심기중 TGF -β1적표체유관。
Objective To explore the protective effect of salvianolate on myocardium in diabetic rats as well as the mechanism of action. Methods 40 male SD rats were randomly divided into model group(n = 30)and control group(n = 10). Intraperitoneal injection of streptozotocin was used for modeling. Successful modeling rats were ran-domly divided into high - dose salvianolate group(n = 10),low - dose salvianolate group(n = 10)and model control group(n = 9). Intraperitoneal injection of streptozotocin lasted for 8 weeks and rats were killed after 10 weeks. Cardiac ultrasound was used to determine cardiac morphology and function indexes. HE staining was used to observe pathomor-phology,and the expression of TGF - β1 was detected by immunohistochemical method. Results Compared with con-trol group,ultrasound indexes decreased significantly,myocardial fiber was in a mess,the expression of TGF - β1 in-creased in model group. Compared with model control group,ultrasound indexes and pathological condition improved, the expression of TGF - β1 decreased,which was more obvious in high - dose group than that in low - dose group(P ﹤0. 05). Conclusion Salvianolate can protect myocardium of diabetic rats,which may relate to decrease expression of TGF - β1 .
