目的:探讨二苯乙烯苷( TSG)对大鼠心肌缺血再灌注损伤的防治作用及其机制。方法用手术线对大鼠冠状动脉左前降支结扎30 min,松开手术线后再灌注120min建立大鼠心肌缺血再灌注损伤模型。将大鼠分为假手术组,缺血再灌注组,缺血再灌注+TSG小剂量(10 mg/kg)干预组,缺血再灌注+TSG大剂量(20 mg/kg)干预组。观察TSG对大鼠心脏形态学、心肌酶学和脂质过氧化及沉默信息调节因子2同源体1(SIRT1)蛋白的影响。结果TSG能改善大鼠心肌组织病理改变,显著降低心肌组织丙二醛( MDA)含量,增强超氧化物歧化酶(SOD)和谷胱甘肽(GSH-Px)的活性,并能够增加心脏组织SIRT1蛋白含量,以大剂量组(20 mg/kg)干预组效果明显。结论TSG对鼠心肌缺血再灌注损伤具有防治作用,其机制可能为上调SIRT1蛋白增强心肌的抗氧化能力。

목적:탐토이분을희감( TSG)대대서심기결혈재관주손상적방치작용급기궤제。방법용수술선대대서관상동맥좌전강지결찰30 min,송개수술선후재관주120min건립대서심기결혈재관주손상모형。장대서분위가수술조,결혈재관주조,결혈재관주+TSG소제량(10 mg/kg)간예조,결혈재관주+TSG대제량(20 mg/kg)간예조。관찰TSG대대서심장형태학、심기매학화지질과양화급침묵신식조절인자2동원체1(SIRT1)단백적영향。결과TSG능개선대서심기조직병리개변,현저강저심기조직병이철( MDA)함량,증강초양화물기화매(SOD)화곡광감태(GSH-Px)적활성,병능구증가심장조직SIRT1단백함량,이대제량조(20 mg/kg)간예조효과명현。결론TSG대서심기결혈재관주손상구유방치작용,기궤제가능위상조SIRT1단백증강심기적항양화능력。
Objective To explore the preventive effect of Tetrahydroxystilbene glucoside ( TSG) on myo-cardia1 ischemia-reperfusion injury in rats .MethodsMyocardial ischemia -reperfusion injury model of rats was estab1ished by ligation of left descending coronary artery for 30 min,then reperfused for 120 min.Rats were di-vided into 4 group:normal control ,ischemia-reperfusion group ,TSG 10 mg/kg group and 20 mg/kg group .The effects of TSG on the histological change ,the cardiac function ,the content of MDA and the expression of SIRT 1 were measured .Results TSG improved the histological change and the cardiac function .TSG also increased the activity of SOD and GSH-Px,and decreased the content of MDA .The decreasing expression of SIRT 1 was re-versed by TSG treatment .Conclusion TSG has protective effect on myocardial ischemia-reperfusion injury by el-evating antioxidative capacity via upregulaiton of SIRT 1 .
