目的:观察芝麻素对AlCl3及D-半乳糖(D-gal)所致大鼠肝损伤的保护作用,并探讨其可能机制。方法:采用AlCl3[15 mg/(kg· d)]灌胃联合D-半乳糖[180 mg/(kg· d)]腹腔注射12周建立大鼠肝损伤模型。各治疗组分别灌胃给予相应药物8周。观察大鼠肝组织病理学改变及胶原纤维增生水平,测定肝组织iNOS表达水平、T-AOC、SOD活性及MDA、NO含量。结果:各治疗组肝细胞损伤和胶原纤维增生较模型组明显减轻,iNOS蛋白表达水平及MDA、NO含量明显下降,T-AOC、SOD活性明显升高(P<00.5或P<0.01)。结论:芝麻素改善AlCl3及D-半乳糖诱导的大鼠肝损伤,其机制可能与提高机体抗氧化能力以及调节iNOS表达有关。

목적:관찰지마소대AlCl3급D-반유당(D-gal)소치대서간손상적보호작용,병탐토기가능궤제。방법:채용AlCl3[15 mg/(kg· d)]관위연합D-반유당[180 mg/(kg· d)]복강주사12주건립대서간손상모형。각치료조분별관위급여상응약물8주。관찰대서간조직병이학개변급효원섬유증생수평,측정간조직iNOS표체수평、T-AOC、SOD활성급MDA、NO함량。결과:각치료조간세포손상화효원섬유증생교모형조명현감경,iNOS단백표체수평급MDA、NO함량명현하강,T-AOC、SOD활성명현승고(P<00.5혹P<0.01)。결론:지마소개선AlCl3급D-반유당유도적대서간손상,기궤제가능여제고궤체항양화능력이급조절iNOS표체유관。
Objective:To observe the protective effect of sesamin on the hepatic injury in rats induced by aluminum chloride ( AlCl3 ) ) and D-galac-tosamine(D -gal) and investigate the potential mechanisms.Methods:Rat models of hepatic injury were established by intragastric administration of AlCl 3 [15 mg/(kg· d)] combined with intraperitoneal injection of D-gal[180 mg/(kg· d)] for 12 weeks,and treated with corresponding drugs for consecu-tive 8 weeks.All rats were examined regarding the pathological changes of the liver tissues and collagen fiber disposition ,and determined concerning the hepatic iNOS expression,activities of T-AOC and SOD as well as levels of MDA and NO.Results:The degree of hepatic injury and collagen fiber disposi-tion were alleviated in rats treated with sesamin,with down-regulated iNOS expression,marked decrease of MDA and NO levels and boosted T-AOC and SOD activities(P<0.05 or P<0.01).Conclusion:Sesamin can improve the hepatic injury in rats induced by AlCl3 and D-gal,and the potential mecha-nisms may be associated with enhancement of antioxidant capacity and down-regulation of iNOS expression.
