目的:评价重组人Ⅱ型肿瘤坏死因子受体-抗体融合蛋白(TNFR-Fc)生物仿制药对大鼠佐剂性关节炎的治疗作用.方法:将大鼠随机分成6组:阴性对照组(生理盐水)、阳性药物组(Enbrel)和 TNFR-Fc 高、中、低质量分数组,空白组(无炎症),每组6只,弗氏完全佐剂(CFA)建立类风湿性关节炎模型.给药13 d后,分别测量大鼠体重、后足足垫厚度,关节炎指数评分,ELISA 法测量细胞因子水平.结果:与阴性对照组相比,Enbrel 组和 TN-FR-Fc 大、中、小质量分数组的大鼠体质量明显增加(P<0.05)、继发性足肿胀减弱(P<0.05)、关节炎评分分值、IL-1β、TNF-α、IL-6含量均减小(P<0.05),IL-10含量升高(P<0.05).结论:TNFR-Fc 通过降低炎性细胞因子IL-1β、IL-6、TNF-α水平以及升高 IL-10水平来发挥治疗类风湿性关节炎的作用.
目的:評價重組人Ⅱ型腫瘤壞死因子受體-抗體融閤蛋白(TNFR-Fc)生物倣製藥對大鼠佐劑性關節炎的治療作用.方法:將大鼠隨機分成6組:陰性對照組(生理鹽水)、暘性藥物組(Enbrel)和 TNFR-Fc 高、中、低質量分數組,空白組(無炎癥),每組6隻,弗氏完全佐劑(CFA)建立類風濕性關節炎模型.給藥13 d後,分彆測量大鼠體重、後足足墊厚度,關節炎指數評分,ELISA 法測量細胞因子水平.結果:與陰性對照組相比,Enbrel 組和 TN-FR-Fc 大、中、小質量分數組的大鼠體質量明顯增加(P<0.05)、繼髮性足腫脹減弱(P<0.05)、關節炎評分分值、IL-1β、TNF-α、IL-6含量均減小(P<0.05),IL-10含量升高(P<0.05).結論:TNFR-Fc 通過降低炎性細胞因子IL-1β、IL-6、TNF-α水平以及升高 IL-10水平來髮揮治療類風濕性關節炎的作用.
목적:평개중조인Ⅱ형종류배사인자수체-항체융합단백(TNFR-Fc)생물방제약대대서좌제성관절염적치료작용.방법:장대서수궤분성6조:음성대조조(생리염수)、양성약물조(Enbrel)화 TNFR-Fc 고、중、저질량분수조,공백조(무염증),매조6지,불씨완전좌제(CFA)건립류풍습성관절염모형.급약13 d후,분별측량대서체중、후족족점후도,관절염지수평분,ELISA 법측량세포인자수평.결과:여음성대조조상비,Enbrel 조화 TN-FR-Fc 대、중、소질량분수조적대서체질량명현증가(P<0.05)、계발성족종창감약(P<0.05)、관절염평분분치、IL-1β、TNF-α、IL-6함량균감소(P<0.05),IL-10함량승고(P<0.05).결론:TNFR-Fc 통과강저염성세포인자IL-1β、IL-6、TNF-α수평이급승고 IL-10수평래발휘치료류풍습성관절염적작용.
Aim:To study the therapeutic effect of rhu TNFR-Fc on adjuvant arthritis and the mecha-nism in rats.Methods:Rats were divided into blank group,control group (normal saline,AA+NS), the Enbrel group and rhu-TNFR-Fc,high,medium and low dose groups.Freunds complete adjuvant (CFA)was used to establish a rat model of rheumatoid arthritis.Weights and paw swelling of the rats were measured,respectively,and the arthritis and viscera index were scored.The cytokine levels were measured by ELISA.Correlations of the testing parameters between groups were statistically analyzed. Results:Compared to the control group,the weights of Enbrel and rhu-TNFR-Fc groups increased signif-icantly (P<0.05 ).However,the paw swelling and the multiple arthritis rating scors were decreased (P<0.05 )in the Enbrel group and rhu-TNFR-Fc with high,medium and low dose group of rats.The con-centrations of plasma IL-1β,TNF-αand IL-6 was found reduced,while the concentration of IL-10 was increased (P<0.05 ).Conclusion:TNFR-Fc fusion protein has a significant therapeutic effect on the rheumatoid arthritis via reducing the plasma concentrations of inflammatory cytokines,such as IL-6, IL-1βand TNF-α.Moreover,TNFR-Fc could also elevate the plasma concentration of IL-10 to suppress inflammation of the synovial cells.
