Broussonetia papyrifera Extract%whitening%tyrosinase activity%melanin synthesis
桑科植物构树的根皮提取物具有较好的美白功效。将90%(体积分数,下同)乙醇提取的构树提取物用适量的去离子水混悬后,上样 HPD-400大孔吸附树脂柱,分别用水、30%乙醇、60%乙醇及95%乙醇洗脱,得Ⅰ、Ⅱ、Ⅲ和Ⅳ四个部位。体外蘑菇酪氨酸酶试验表明,Ⅰ、Ⅱ、Ⅲ和Ⅳ四个部位均具有一定的酪氨酸酶活性抑制作用,其中部位Ⅲ的抑制作用最强,其IC50值为12.5μg/mL。在B16细胞中,相同浓度下部位Ⅲ表现出最强的抑制酪氨酸酶活性和抑制黑色素合成的作用。因此,构树提取物通过 HPD-400大孔吸附树脂得到的部位Ⅲ成分,可作为美白原料用于化妆品的开发。

The root and bark extract of Broussonetia papyrifera,a moraceous plant,was reported to provide a good activity in whitening. In this study,90% ethanol extract of Broussonetia papyrifera was suspended by deionized water and loaded to the column of macroporous resin HPD-400. Then the column was washed with water,30% ethanol,60% ethanol and 95%ethanol in turn to give I,II,III and IV four parts of extract. The result of mushroom tyrosinase experiment demonstrated that I,II,III and IV all showed inhibition activity to tyrosinase to some extent,of which III provided the strongest inhibition with an IC50 of 12.5μg/mL. In B16 cells,III also showed the strongest inhibition to tyrosinase activity and melanin synthesis at the same concentration,compared with I,II and IV. Part III of Broussonetia papyrifera extract,separated by macroporous resin HPD-400,may be used as whitening raw material for cosmetics.
