采用线粒体COI和Cyt b基因序列分析相结合的方法,对伊犁河和塔里木河水系6个斑重唇鱼( Diptychus maculates)群体遗传差异和分化水平进行了比较研究。 AMOVA分析结果显示,斑重唇鱼群体间遗传差异显著大于群体内,群体平均遗传分化指数ΦST分别为0.9802( COI)和0.8863( Cyt b),除伊犁河水系两群体差异不显著外( P>0.05),其他两两群体间遗传差异均显著。基于Kimura双参数法计算群体间平均遗传距离发现,不仅两水系间群体存在差异,塔里木河水系内部群体也产生了较大的遗传分化。单倍型网络图和系统关系树显示库玛拉克河群体存在两组不同单倍型类群,分别与伊犁河和塔里木河水系单倍型聚类。根据分子钟假说,基于Cyt b基因序列推算斑重唇鱼两支系发生分歧的时间大约在2.41 Ma年的上新世末到更新世初。天山第四纪构造运动第一幕可能是两水系斑重唇鱼发生遗传分化的重要原因,而塔里木河水系群体发生遗传分化可能与塔克拉玛干沙漠的阻隔有关。

In this study, the mitochondrial COI and Cyt b gene sequences were analyzed to reveal the genetic diversity and divergence level in six Diptychus maculatus populations .AMOVA analysis showed that genetic variation among populations was significantly greater than that within populations , and the average genetic differentiation index ΦST were 0.980 2 ( COI) and 0.886 3 ( Cyt b) respectively.Genetic differences between two populations were significant except two popula-tions from Ili River ( P>0.05 ).Average genetic distance based on Kimura two-parameter suggested that not only popula-tions between two river systems showed significant genetic differentiations but certain populations among Tarim River also showed differentiations.Haplotype network charts and phylogenetic trees both revealed Kumalake River population existed two different haplogroups.According to the molecular clock hypothesis , phylogenetic divergence time was about 2.41 Ma when it was late Pliocene to early Pleistocene.The Quaternary tectonic movement in the first act of Tianshan Mountain might be the most important reason for genetic divergence in D.maculates populations from two water systems.However , the ge-netic divergence in D.maculates populations from Tarim River might be caused by the separation of Taklimakan Desert.
