以茄门甜椒种子为试材,对无菌苗培养前的种子消毒剂种类及不同消毒时间、浓度等的消毒效果进行了比较研究;对影响无菌苗生长的培养基配方、pH值及生长时间等因素进行了探讨。结果表明,甜椒无菌苗的最佳消毒流程和生长条件为:55℃无菌水浸种15 min,室温静置4~5 h,用10%NaClO消毒30 min,无菌水冲洗3次,接种至1/2MS培养基+15 g/L蔗糖+7.5 g/L琼脂,pH值为5.8,(25±1)℃,子叶长出6~10 d左右,即可作为转基因用无菌苗。
以茄門甜椒種子為試材,對無菌苗培養前的種子消毒劑種類及不同消毒時間、濃度等的消毒效果進行瞭比較研究;對影響無菌苗生長的培養基配方、pH值及生長時間等因素進行瞭探討。結果錶明,甜椒無菌苗的最佳消毒流程和生長條件為:55℃無菌水浸種15 min,室溫靜置4~5 h,用10%NaClO消毒30 min,無菌水遲洗3次,接種至1/2MS培養基+15 g/L蔗糖+7.5 g/L瓊脂,pH值為5.8,(25±1)℃,子葉長齣6~10 d左右,即可作為轉基因用無菌苗。
이가문첨초충자위시재,대무균묘배양전적충자소독제충류급불동소독시간、농도등적소독효과진행료비교연구;대영향무균묘생장적배양기배방、pH치급생장시간등인소진행료탐토。결과표명,첨초무균묘적최가소독류정화생장조건위:55℃무균수침충15 min,실온정치4~5 h,용10%NaClO소독30 min,무균수충세3차,접충지1/2MS배양기+15 g/L자당+7.5 g/L경지,pH치위5.8,(25±1)℃,자협장출6~10 d좌우,즉가작위전기인용무균묘。
Using Qiemen sweet pepper as materials , the disinfection effects of different disinfectant a-gents, disinfection times and contents on seeds before culture were compared , and the effects of medium for-mula, pH value and growth time on growth of aseptic seedling were investigated .The results showed that the optimum disinfection method and culture conditions for aseptic sweet pepper seedling were soaking seeds in a -septic water of 55℃for 15 minutes, then staying at room temperature for 4~5 hours, disinfecting with 10%NaClO for 30 minutes, washing 3 times with aseptic water and then culturing in medium of 1/2MS+15 g/L sugar+7.5 g/L agar with pH value of 5.8 under (25 ±1)℃.When the cotyledon sprouted for 6~10 days, the seedlings could be used for genetic transformation .
