本研究采用不同浓度24-表油菜素内酯处理海洋红藻龙须菜,研究热激胁迫后龙须菜的生长速率、叶绿素荧光参数、抗氧化酶活性、甘露醇、丙二醛、藻胆蛋白含量,以及 HSP70、PE、GR 基因的相对表达水平变化规律。结果显示,0.1 mg/L 24-表油菜素内酯的处理组生长速率最大,显著高于其他处理组及对照组;叶绿素荧光参数 Fv/Fm、qP 和ΦPS Ⅱ均随高温逆境时间延长呈现下降趋势,荧光参数NPQ 随着高温逆境时间延长呈现先上升后下降的趋势,且处理组各项荧光参数值均高于对照组;处理组的抗氧化酶(SOD 和 POD)的酶活显著高于对照组;甘露醇及藻胆蛋白含量均在0.1 mg/L 处理组最高,在第三天达到最大值,而丙二醛含量则在0.1 mg/L 处理组最低,在第三天达到最小值;处理组 HSP70、PE、GR 基因的相对表达量均高于对照组,且0.1 mg/L 处理组各基因表达量最高。以上结果表明24-表油菜素内酯能提高龙须菜的抗高温能力,且0.1 mg/L 处理组效果最好。

본연구채용불동농도24-표유채소내지처리해양홍조룡수채,연구열격협박후룡수채적생장속솔、협록소형광삼수、항양화매활성、감로순、병이철、조담단백함량,이급 HSP70、PE、GR 기인적상대표체수평변화규률。결과현시,0.1 mg/L 24-표유채소내지적처리조생장속솔최대,현저고우기타처리조급대조조;협록소형광삼수 Fv/Fm、qP 화ΦPS Ⅱ균수고온역경시간연장정현하강추세,형광삼수NPQ 수착고온역경시간연장정현선상승후하강적추세,차처리조각항형광삼수치균고우대조조;처리조적항양화매(SOD 화 POD)적매활현저고우대조조;감로순급조담단백함량균재0.1 mg/L 처리조최고,재제삼천체도최대치,이병이철함량칙재0.1 mg/L 처리조최저,재제삼천체도최소치;처리조 HSP70、PE、GR 기인적상대표체량균고우대조조,차0.1 mg/L 처리조각기인표체량최고。이상결과표명24-표유채소내지능제고룡수채적항고온능력,차0.1 mg/L 처리조효과최호。
To explore the effect of 24-epibrassinolide on the marine red algae Gracilaria lemaneiformis under high temperature stress,the growth rate,chlorophyll fluorescence parameters,antioxidant enzyme activity,as well as the contains of mannitol,malondialdehyde,phycobiliprotein,and the relative expression levels of HSP70 ,PE and GR were detected in G.lemaneiformis after treated with different concentration of 24-epibrassinolide and heat shock stress.Results showed that the growth rate of 0.1 mg/L treatment group was significantly higher than that of other groups.Chlorophyll fluorescence parameters of Fv/Fm,qP and ΦPS Ⅱ decreased while parameters of NPQ first increased and then decreased along with the extension of high temperature stress.Chlorophyll fluorescence pa-rameters in all treatment groups were higher than those in the control groups.The activities of SOD and POD in treatment groups were significantly higher than those in the control groups.The detected mannitol and phycobilip-rotein levels were the highest in 0.1 mg/L treatment group,and increased to the summit at 3 d post treatment. The detected malondialdehyde level was the lowest in 0.1 mg/L treatment group,and decreased to the bottom at 3 d post treatment.The relative gene expression levels of HSP70,PE and GR in treatment groups were higher than those in the control groups.They all presented the highest in 0.1 mg/L treatment group.The results indicated that 24-epibrassinolide could improve high temperature resistance of G.lemaneiformis,and the 0.1 mg/L treat-ment group showed the best effect.
