目的:观察蓬草除痹汤( PCCB)对Ⅱ型胶原诱导关节炎( CIA)大鼠模型外周血中细胞因子IL-17、IL-1β、TNF-α及T细胞亚群CD4+T、CD8+T、CD4+T/CD8+T的干预作用,探讨PCCB治疗类风湿关节炎( RA)的抗炎免疫作用机制。方法建立CIA大鼠模型,随机分为PCCB组、雷公藤组、空白组及模型组,连续给药28 d,采用酶联免疫吸附( ELISA)法、流式细胞术评价PCCB对CIA大鼠关节炎指数(AI)及大鼠血清IL-17、IL-1β、TNF-α和CD4+T、CD8+T、CD4+T/CD8+T表达的影响。结果治疗28 d后,PCCB组、雷公藤组关节肿胀度及AI显著改善(P均<0.01);血清IL-17、IL-1β、TNF-α水平明显降低(P均<0.01);CD4+T降低,CD8+T升高,CD4+T/CD8+T显著降低,且PCCB组的抑制作用明显优于雷公藤组(P<0.05)。结论 PCCB能有效改善CIA大鼠关节炎指数,下调大鼠血清中致炎因子IL-17、IL-1β、TNF-α的水平,调节大鼠血清中CD4+T、CD8+T及CD4+T/CD8+T 比例,这可能是PCCB治疗RA的主要机制,为临床应用及进一步研究奠定了基础。

목적:관찰봉초제비탕( PCCB)대Ⅱ형효원유도관절염( CIA)대서모형외주혈중세포인자IL-17、IL-1β、TNF-α급T세포아군CD4+T、CD8+T、CD4+T/CD8+T적간예작용,탐토PCCB치료류풍습관절염( RA)적항염면역작용궤제。방법건립CIA대서모형,수궤분위PCCB조、뢰공등조、공백조급모형조,련속급약28 d,채용매련면역흡부( ELISA)법、류식세포술평개PCCB대CIA대서관절염지수(AI)급대서혈청IL-17、IL-1β、TNF-α화CD4+T、CD8+T、CD4+T/CD8+T표체적영향。결과치료28 d후,PCCB조、뢰공등조관절종창도급AI현저개선(P균<0.01);혈청IL-17、IL-1β、TNF-α수평명현강저(P균<0.01);CD4+T강저,CD8+T승고,CD4+T/CD8+T현저강저,차PCCB조적억제작용명현우우뢰공등조(P<0.05)。결론 PCCB능유효개선CIA대서관절염지수,하조대서혈청중치염인자IL-17、IL-1β、TNF-α적수평,조절대서혈청중CD4+T、CD8+T급CD4+T/CD8+T 비례,저가능시PCCB치료RA적주요궤제,위림상응용급진일보연구전정료기출。
Objective It is to observe the intervention effect on cytokine of IL -17 , IL-1β, TNF-αand T-lymphocyte subset of CD4 +T,CD8 +T and CD4 +T/CD8 +T in peripheral blood of rats with collagen -induced arthritis Ⅱ( CIA) treated by the Peng Cao rheumatism -syndrome eliminating decoction ( PCCB, abbreviation for the decoction's Chinese name: Peng Cao Chu Bi ) , and to explore its mechanism of immune anti -inflammatory action .Methods CIA rat models were established and randomly divided them into 4 groups:PCCB group , tripterygium glycosides group , blank group and model group , and all the groups were treated for 28 days.Enzyme-linked immunosorbent assay (ELISA) and flow cytometry were used to evaluate the effects on arthritis index ( AI) and on the expression levels of IL -17 , IL-1βand TNF-αand T-lymphocyte subset of CD4 +T,CD8 +T and CD4 +T/CD8 +T in serum of CIA rats treated by PCCB .Results After 28 days'treatment, compared with model group, the AI of the rats in the PCCB and TG groups were significantly improved (P<0.01), the levels of IL-17, IL-1βand TNF-αin serum were reduced (P<0.01), CD4 +were reduced, CD8 +increased (P<0.01) and CD4 +T/CD8 +T reduced obviously (P<0.05), while the inhibiting effect in PCCB group were superior obviously to that in TG group (P<0. 05).Conclusion PCCB could effectively improve the AI of CIA rats , reduce the level of pro-inflammatory cytokine of IL -1, IL-1βand in serum, regulate the ratio of CD4 +T,CD8 +T and CD4 +T/CD8 +T in serum of the rats.That might suggest a key mechanism to remedy RA by PCCB , which may lay a foundation of clinical application for RA treatment .
