目的:探讨白细胞介素24(IL-24)对成人急性白血病骨髓单个核细胞(BMMNC)体外生长的影响,并初步探讨其作用的可能机制。方法:随机选择在石河子大学一附院住院的急性白血病患者15例,均经FAB分型或免疫学分型确诊,其中确诊未治的成人急性淋巴细胞白血病(ALL)患者5例,成人急性非淋巴细胞白血病(ANLL)患者10例。取患者骨髓,分离单个核细胞,以不同浓度IL-24作用于成人急性白血病BMMNC。应用四氮唑蓝(MTT)比色还原法,检测IL-24对急性白血病BMMNC体外生长的影响;采用AnnexinV-FITC染色流式细胞术,观察检测IL-24对急性白血病BMMNC增殖状况的影响;利用逆转录-聚合酶链反应(RT-PCR),检测IL-24对急性白血病BMMNC中caspase-3和bcl-2表达的影响。结果:MTT法发现IL-24对体外培养的白血病BMMNC有明显的生长抑制作用,且呈时间、剂量依赖性,与阴性对照组相比差异有显著性(P<0.05)。IL-24浓度50 ng/mL作用于急性白血病BMMNC 48 h后,Annexin V-PI检测结果显示凋亡率为(39.21±5.79)%,高于对照组(P<0.01)。 RT-PCR检测显示,不同浓度IL-24处理白血病BMMNC,能够增加caspase-3 mRNA的表达,降低bcl-2 mRNA的表达。结论:IL-24对体外培养的成人急性白血病BMMNC,有明显的生长抑制、诱导凋亡作用。 IL-24可以上调caspase-3 mRNA表达,下调bcl-2 mRNA表达,可能是IL-24抑制急性白血病BMMNC生长作用的重要机制。

목적:탐토백세포개소24(IL-24)대성인급성백혈병골수단개핵세포(BMMNC)체외생장적영향,병초보탐토기작용적가능궤제。방법:수궤선택재석하자대학일부원주원적급성백혈병환자15례,균경FAB분형혹면역학분형학진,기중학진미치적성인급성림파세포백혈병(ALL)환자5례,성인급성비림파세포백혈병(ANLL)환자10례。취환자골수,분리단개핵세포,이불동농도IL-24작용우성인급성백혈병BMMNC。응용사담서람(MTT)비색환원법,검측IL-24대급성백혈병BMMNC체외생장적영향;채용AnnexinV-FITC염색류식세포술,관찰검측IL-24대급성백혈병BMMNC증식상황적영향;이용역전록-취합매련반응(RT-PCR),검측IL-24대급성백혈병BMMNC중caspase-3화bcl-2표체적영향。결과:MTT법발현IL-24대체외배양적백혈병BMMNC유명현적생장억제작용,차정시간、제량의뢰성,여음성대조조상비차이유현저성(P<0.05)。IL-24농도50 ng/mL작용우급성백혈병BMMNC 48 h후,Annexin V-PI검측결과현시조망솔위(39.21±5.79)%,고우대조조(P<0.01)。 RT-PCR검측현시,불동농도IL-24처리백혈병BMMNC,능구증가caspase-3 mRNA적표체,강저bcl-2 mRNA적표체。결론:IL-24대체외배양적성인급성백혈병BMMNC,유명현적생장억제、유도조망작용。 IL-24가이상조caspase-3 mRNA표체,하조bcl-2 mRNA표체,가능시IL-24억제급성백혈병BMMNC생장작용적중요궤제。
Objective The aim of this study was to observe the effects of interleukin-24 (IL-24) on the growth of acute leukemin bone marrow mononuclear cells (BMMNC) in vitro. And to investigate the mechanisms of IL-24 on acute leukemin BMMNC. Methods The group of acute lymphocytic leukemia (ALL) had 5 adults, the group of acute myeloid leukemia (ANLL) had 10 adults who did not receive any therapy. The acute leukemin BMMNC were separated from bone marrow. BMMNC were treated with IL-24 in vitro. The effects of IL-24 on acute leukemin BMMNC in vitro was detected by MTT assay. Apoptosis of acute leukemin BMMNC was measured by flow cytometry with AnnexinV-FITC staining. The expression of caspase-3 and bcl-2 in acute leukemin BMMNC was detected by RT-PCR. Results IL-24 can significantly inhibit the proliferation of acute leukemia BMMNC which was detected by MTT assay, and its effects were dose-dependent and time-dependent. After acute leukemia BMMNC were exposed to IL-24 for 48 h, the apoptotic rate of the group treated with IL-24 of 50 ng/mL was increased obviously. IL-24 increased the mRNA expression level of caspase-3, decreased the mRNA expression level of bcl-2 in acute leukemia BMMNC. Conclusion IL-24 can suppress the growth and induce apoptosis of acute leukemia BMMNC in vitro. IL-24 may inhibit the growth of acute leukemia BMMNC by up-regulating the expression of caspase-3 and down-regulating the expression of bcl-2.
