目的:开展芳冰鼻吸剂对大鼠镇静助眠的药效研究,评价其可能的作用机理。方法:采用改良多平台水环境法造模。观察大鼠造模前后行为、体重、血常规等变化,并采用ELISA测定给药后大鼠血清、海马区5-HT、DA、NA的含量变化。结果:睡眠剥夺后给予药物治疗,低剂量组可显著缩短大鼠入睡时间,同时与环境对照组比较,造模后大鼠血清中的5-HT 及海马中的DA含量均降低,中剂量组血清中DA及NA的含量明显低于阳性对照组;与模型组比较,环境对照组、阳性对照组和高剂量组海马中DA的含量显著增加。结论:芳冰鼻吸剂具有较好的镇静助眠的疗效,其作用机理主要是通过升高大鼠血清中5-HT、NA的含量,降低海马中NA含量来达到镇静助眠的功效。

목적:개전방빙비흡제대대서진정조면적약효연구,평개기가능적작용궤리。방법:채용개량다평태수배경법조모。관찰대서조모전후행위、체중、혈상규등변화,병채용ELISA측정급약후대서혈청、해마구5-HT、DA、NA적함량변화。결과:수면박탈후급여약물치료,저제량조가현저축단대서입수시간,동시여배경대조조비교,조모후대서혈청중적5-HT 급해마중적DA함량균강저,중제량조혈청중DA급NA적함량명현저우양성대조조;여모형조비교,배경대조조、양성대조조화고제량조해마중DA적함량현저증가。결론:방빙비흡제구유교호적진정조면적료효,기작용궤리주요시통과승고대서혈청중5-HT、NA적함량,강저해마중NA함량래체도진정조면적공효。
Objective:To investigate the effects of Fangbing nasal inhalant on sedative good sleep in rats and evaluate its potential effective theory. Methods:The sleep deprivation model of rats was established through modified multiple platform method. The conducts of rats, the weight of rats and the blood routine analysis were measured before and after model. At the same time, the content of 5-HT, DA, NA both in serum and hippocampus of rats were tested by ELISA method after drug treatment. Results: After sleep deprivation, as compared with control group, the content of WBC and W-SCC in normal control group increased significantly, but the content of PLT, W-MCC and W-LCC decreased significantly. After drug therapy, the time to fall asleep in rats in FB low dose group was significantly shortened. In the meantime, after model, both 5-HT content in serum and DA content in hippocampus decreased when compared with the environmental control group. The content of DA and NA in serum was significantly lower in middle dose group than that in the positive control group. In terms of DA content in hippocampus, there was statistical significance between model group rats and other groups. Conclusion:Fangbing nasal inhalant has a good effectiveness for sedative sleep, which effect mechanism is related to the increasing content of 5-HT and NA in serum and decreasing NA content in hippocampus to achieve the efficacy of sedative good sleep.
