目的:研究山竹果肉提取物中多糖、蛋白质含量及多糖中单糖组成和山竹果皮提取物中总黄酮的含量。方法取山竹果皮用95%乙醇回流提取,减压浓缩得到山竹果皮提取物;山竹果肉分别用95%乙醇,蒸馏水回流提取,合并蒸馏水提取液减压浓缩得到山竹果肉提取物。采用可见-紫外分光光度法以苯酚-硫酸、考马斯亮蓝显色分别测定山竹果肉提取物多糖和蛋白质的含量;亚硝酸钠-硝酸铝显色测定山竹果皮提取物总黄酮的含量;果肉提取物多糖水解产物进行甲基化反应,以气相色谱检测多糖水解产物。结果分别采用上述方法能够分别准确测定出山竹果皮提取物中总黄酮含量为6.30%,果肉提取物中多糖和蛋白质含量分别为84.99%和1.64%,确定果肉提取物多糖水解产物中的4种单糖分别为院D-葡萄糖,D-果糖,D-阿拉伯糖,D-木糖,其摩尔比为1.34:1.66:1.00:40.06。结论本研究建立的含量测定方法操作简单,重复性好,结果可靠,可用于山竹提取物的质量控制。

목적:연구산죽과육제취물중다당、단백질함량급다당중단당조성화산죽과피제취물중총황동적함량。방법취산죽과피용95%을순회류제취,감압농축득도산죽과피제취물;산죽과육분별용95%을순,증류수회류제취,합병증류수제취액감압농축득도산죽과육제취물。채용가견-자외분광광도법이분분-류산、고마사량람현색분별측정산죽과육제취물다당화단백질적함량;아초산납-초산려현색측정산죽과피제취물총황동적함량;과육제취물다당수해산물진행갑기화반응,이기상색보검측다당수해산물。결과분별채용상술방법능구분별준학측정출산죽과피제취물중총황동함량위6.30%,과육제취물중다당화단백질함량분별위84.99%화1.64%,학정과육제취물다당수해산물중적4충단당분별위원D-포도당,D-과당,D-아랍백당,D-목당,기마이비위1.34:1.66:1.00:40.06。결론본연구건립적함량측정방법조작간단,중복성호,결과가고,가용우산죽제취물적질량공제。
Objective To study on the content of polysaccharides, proteins, flavonoids and the monosaccharides consti-tution from Garcinia (G.) mangostana extract. Methods The pericarp was extracted with 95%ethanol for reflux, and the pericarp extractive of G. mangostana was obtained with decompress and concentration;the sarcocarp was extracted with 95% ethanol for reflux and was distilled by water, the sarcocarp extractive of G. mangostana was obtained with decom-press and concentration by the distilled water extract. The ultraviolet-visible spectrophotometric method was used for determined content, the polysaccharides content of the pericarp extractive was determined by the phenol-sulfuric acid color reaction method, the proteins content was determined of the pericarp extractive by the coomassie brilliant blue method, and the sodium nitrate-nitric acid aluminum method was used to determine the content of flavonoids by the sarcocarp extractive. G. mangostana polysaccharides of monosaccharides constitution with methylation was determined by using hydrolysis and gas chromatography. Results The above methods were used to accurate determine constituents content of G. mangostana. The content of total flavonoids in the extracts was 6.30%of the skin, the fruit extract polysac-charide and protein content were 84.99% and 1.64% respectively from G. mangostana. Four monosaccharides were de-termined by gas chromatography, they were D-glucose, D-fructose, D-arabinose and D-xylose in G. mangostana, the molar ratio was 1.34:1.66:1.00:40.06. Conclusion The establishing operation method in this study of the content determi-nation is simple, highly reproducible and reliable, it can be used for the quality control in the extract of G. mangostana.
