该文以琼岛杨为研究对象,研究外植体、消毒方法,以及植物激素对琼岛杨愈伤组织的诱导、增殖和分化的影响。研究结果表明:琼岛杨以叶片作为外植体,0.1%的升汞消毒6min.,无菌水冲洗5次比较理想。激素配比6-BA 0.5mg/L+2-4D 1.0mg/L 能更好地诱导和促进愈伤组织的生长,增值率达到了80%,诱导的愈伤组织呈现翠绿色,颗粒饱满;激素配比6-BA 2.0mg/L+ IBA 0.5mg/L 有利于愈伤组织分化出带绒毛的芽点。

The article studies the effect of the explant, disinfection method, and phytohormone on the callus in-duction, reproduction and differentiation of Populus qiongdaoensis T. Hong et P. Luo. The results showed that it was a better way using the leaf as explant sterilized by 0.1% (w/v) aqueous mercuric chloride solution for 6 min with sterile water washing for 5 times. The combination of 6-BA 0.5 mg/L and 2-4 d 1.0 mg/L would be better to promote the growth of callus, and the induction rate of reproduction reached 80%. The induction of callus presented emerald and starlatine. The ratio of phytohormone supplemented 6 - BA 2.0 mg/L with IBA 0.5 mg/L was conducive to the callus differentiate into bud points with fluff.
