目的:探讨骨髓间充质干细胞移植对脑出血大鼠的行为和血肿周围神经细胞凋亡的影响。方法全骨髓贴壁法分离培养大鼠骨髓间充质干细胞,使用立体定向纹状体注入胶原酶法制作大鼠脑出血模型。 SD大鼠30只,随机分为对照组和移植组(各15只),对照组制作脑出血模型,不移植;移植组制作脑出血模型并于造模后48h经立体定向脑内注射1×105个干细胞,并于移植后1d、3d、5d、7d、14d进行神经功能缺损评分,对照组在相应时间点同样评分,各组再根据时间的不同随机分成五个亚组(每组3只),在相应时间点处死大鼠行免疫组织化学法检测细胞凋亡。结果在移植后1 d、3 d,两组大鼠评分及血肿周围脑组织内凋亡细胞的数量均较高,两组之间无显著性差异。在移植后第5 d、7 d,两组大鼠评分及血肿周围脑组织内凋亡细胞的数量开始下降,移植组下降幅度大于对照组,两组之间差异显著。移植后14 d,两组之间无显著性差异。结论骨髓间充质干细胞移植可明显促进大鼠脑出血后神经功能的恢复,其机制可能与下调血肿周围神经细胞凋亡有关。
目的:探討骨髓間充質榦細胞移植對腦齣血大鼠的行為和血腫週圍神經細胞凋亡的影響。方法全骨髓貼壁法分離培養大鼠骨髓間充質榦細胞,使用立體定嚮紋狀體註入膠原酶法製作大鼠腦齣血模型。 SD大鼠30隻,隨機分為對照組和移植組(各15隻),對照組製作腦齣血模型,不移植;移植組製作腦齣血模型併于造模後48h經立體定嚮腦內註射1×105箇榦細胞,併于移植後1d、3d、5d、7d、14d進行神經功能缺損評分,對照組在相應時間點同樣評分,各組再根據時間的不同隨機分成五箇亞組(每組3隻),在相應時間點處死大鼠行免疫組織化學法檢測細胞凋亡。結果在移植後1 d、3 d,兩組大鼠評分及血腫週圍腦組織內凋亡細胞的數量均較高,兩組之間無顯著性差異。在移植後第5 d、7 d,兩組大鼠評分及血腫週圍腦組織內凋亡細胞的數量開始下降,移植組下降幅度大于對照組,兩組之間差異顯著。移植後14 d,兩組之間無顯著性差異。結論骨髓間充質榦細胞移植可明顯促進大鼠腦齣血後神經功能的恢複,其機製可能與下調血腫週圍神經細胞凋亡有關。
목적:탐토골수간충질간세포이식대뇌출혈대서적행위화혈종주위신경세포조망적영향。방법전골수첩벽법분리배양대서골수간충질간세포,사용입체정향문상체주입효원매법제작대서뇌출혈모형。 SD대서30지,수궤분위대조조화이식조(각15지),대조조제작뇌출혈모형,불이식;이식조제작뇌출혈모형병우조모후48h경입체정향뇌내주사1×105개간세포,병우이식후1d、3d、5d、7d、14d진행신경공능결손평분,대조조재상응시간점동양평분,각조재근거시간적불동수궤분성오개아조(매조3지),재상응시간점처사대서행면역조직화학법검측세포조망。결과재이식후1 d、3 d,량조대서평분급혈종주위뇌조직내조망세포적수량균교고,량조지간무현저성차이。재이식후제5 d、7 d,량조대서평분급혈종주위뇌조직내조망세포적수량개시하강,이식조하강폭도대우대조조,량조지간차이현저。이식후14 d,량조지간무현저성차이。결론골수간충질간세포이식가명현촉진대서뇌출혈후신경공능적회복,기궤제가능여하조혈종주위신경세포조망유관。
Objective To explore the effect of the behavioral test and neurological cell apoptosis on transplantation of bone mesenchymal stem cells (BMSCs) for experimental intracerebral hemorrhage (ICH) in rats.Methods BMSCs were isolated and cultured using the whole bone marrow adherent method.A rat model of ICH was established by stereotaxical injection of collagenase Ⅰinto the right caudate nucleus .30 Sprague-Dawley ( SD ) rats were randomly divided into control group ( 15 ) and transplantation group (15).The control group were injected with collagenaseⅠonly,the transplant-ation group were transfused with BMSCs (1 ×105/rat) at 48 hours after surgery .On 1,3,5,7,14 days afer transplantation neurological function tests were performed among all groups and animals were sacrificed at the corresponding time points for detecting the neurological cell apoptosis .Results On 1,3 days after transplantation ,neurological function tests score and neurological cell apoptosis of two groups are not different .On 5,7 days after transplantation ,neurological function tests score and neurological cell apoptosis of two groups are significantly different .On 14 days after transplantation , neurological function tests score and neurological cell apoptosis of two groups have no difference .Conclusion Transplantation of BMSCs could significantly promote the neurological function recovery of ICH rats , which might be associated with its downregulation of neurological cell apoptosis .
