目的:探讨补阳还五汤对脑缺血后大鼠脑组织NF-κB/p50表达的影响。方法:将180只SD大鼠随机分为空白组、假手术组、模型组、补阳还五汤组、PDTC(呲咯烷二硫代氨基甲酸盐)组和米诺环素组,各30只,每组按给药后7天、14天、21天3个时间点再各分为3个亚组,观察各组不同时间点的神经功能评分,免疫组化检测缺血脑组织NF-κB/p50蛋白表达情况。结果:补阳还五汤组、米诺环素组、PDTC组各时间点神经功能缺失症状评分和脑组织NF-κB/p50阳性细胞数与模型组比较,均明显减少(P<0.05或P<0.01);用药7天、14天后,补阳还五汤组NF-κB/p50阳性细胞数与米诺环素组,PDTC组组间差异无统计学意义(P>0.05),用药21天后,补阳还五汤组与PDTC组比较,具有显著性差异(P<0.05)。结论:补阳还五汤可促进脑缺血损伤后神经功能恢复,调控缺血脑组织炎症因子NF-κB/p50的表达可能是其治疗脑缺血的作用机制之一。
目的:探討補暘還五湯對腦缺血後大鼠腦組織NF-κB/p50錶達的影響。方法:將180隻SD大鼠隨機分為空白組、假手術組、模型組、補暘還五湯組、PDTC(呲咯烷二硫代氨基甲痠鹽)組和米諾環素組,各30隻,每組按給藥後7天、14天、21天3箇時間點再各分為3箇亞組,觀察各組不同時間點的神經功能評分,免疫組化檢測缺血腦組織NF-κB/p50蛋白錶達情況。結果:補暘還五湯組、米諾環素組、PDTC組各時間點神經功能缺失癥狀評分和腦組織NF-κB/p50暘性細胞數與模型組比較,均明顯減少(P<0.05或P<0.01);用藥7天、14天後,補暘還五湯組NF-κB/p50暘性細胞數與米諾環素組,PDTC組組間差異無統計學意義(P>0.05),用藥21天後,補暘還五湯組與PDTC組比較,具有顯著性差異(P<0.05)。結論:補暘還五湯可促進腦缺血損傷後神經功能恢複,調控缺血腦組織炎癥因子NF-κB/p50的錶達可能是其治療腦缺血的作用機製之一。
목적:탐토보양환오탕대뇌결혈후대서뇌조직NF-κB/p50표체적영향。방법:장180지SD대서수궤분위공백조、가수술조、모형조、보양환오탕조、PDTC(자각완이류대안기갑산염)조화미낙배소조,각30지,매조안급약후7천、14천、21천3개시간점재각분위3개아조,관찰각조불동시간점적신경공능평분,면역조화검측결혈뇌조직NF-κB/p50단백표체정황。결과:보양환오탕조、미낙배소조、PDTC조각시간점신경공능결실증상평분화뇌조직NF-κB/p50양성세포수여모형조비교,균명현감소(P<0.05혹P<0.01);용약7천、14천후,보양환오탕조NF-κB/p50양성세포수여미낙배소조,PDTC조조간차이무통계학의의(P>0.05),용약21천후,보양환오탕조여PDTC조비교,구유현저성차이(P<0.05)。결론:보양환오탕가촉진뇌결혈손상후신경공능회복,조공결혈뇌조직염증인자NF-κB/p50적표체가능시기치료뇌결혈적작용궤제지일。
Objective:To observe the effect of Buyang Huanwu decoction on NF -κB/p50 expression in brain tissue of rats with focal cerebral ischemia injury .Methods:One hundred and eighty SD rats were randomly di-vided into normal group , Sham -operated group , ischemia group , BYHWD treatment group , PDTC treatment group and MC treatment group .Each group was divided into three subgroups according to the time after admin-istration for 7d,14d,21d respectively.Neurological function score and NF -κB/p50 protein levels were exam-ined by using immunohistochemistry in each group at different time points .Results:Neurological function score and expression of NF-κB/p50 in BYHWD,MC,PDTC treatment group were significantly decreased compared with those in model group at the same time (P<0.05 or P<0.01);after 7,14 days’treatment, the difference of the expression of NF -κB/p50 was not significant ( P>0.05) .Between the treatment groups ,after 21 days’ treatment BYHWD displayed the trifle superiority in comparison with PDTC ( P<0 .05 ) .Conclusion:Buyang Huanwu decoction can improve the recovery of cerebral ischemic neurological injury and regulating cerebral in -flammatory cytokine NF-κB/p50 expression of ischemia rats may be one of its therapeutic mechanisms for cerebral ischemia .
