目的:建立贝母花的定性鉴别和含量测定方法。方法:采用薄层色谱法进行定性鉴别;ELSD-HPLC法测定贝母素甲和贝母素乙的含量,采用Agilent 1260高效液相色谱仪, Agilent 380蒸发光散射检测器检测,C18色谱柱(5μm,4.6mm ×150mm,);流动相:乙腈-水-三乙胺(68∶32∶0.03),流速:1.0 mL/min;柱温:30℃;漂移管温度:90℃;进样量:20μL。结果:薄层色谱专属性强;贝母素甲、贝母素乙的线性范围分别是0.0185~0.185mg/mL(r=0.9998),0.0188~0.188mg/mL(r=0.9997);平均加样回收率为100.40%,RSD为1.50%(n=6)。结论:该方法简便、准确,重现性好,可作为控制贝母花质量的方法。

To establish the quality standard for flowers of Fritillaria thunbergii Miq.Methods:Flowers of Fritillaria thunbergii Miq.were identified by TLC .The contents of peimine and peiminninne were determined by ELSD-HPLC:The optimal conditions of separation and detection were achieved on a C18 ana-lytical column with an mobile phase consisting of acetonitrile -water -triethylamine ( 68∶32∶0 .03 ) at the flow-rate of 1.0 mL/min;the temperature for the detector drift tube was set at 90℃and the nitrogen flow-rate was at 2.2L/min, 20μL as the injection volume .Results:The identification by TLC was highly specific . The calibration curves for peimine and peiminine were linear over the range of 0 .0185~0 .185 mg/mL (r=0.9998 ) and 0.0188~0.188 mg/mL(r=0.9997) , respectively .The average recovery rate was 100.40%with RSD of 1.50%(n=6).Conclusion:The method is simple and reproducible . It can serve as a quantita-tive determination and quality control of flowers of Fritillaria thunbergii Miq.
