目的:研究二氢槲皮素对H2 O2诱导H9c2心肌细胞损伤的保护作用。方法:建立H2 O2诱导H9c2心肌细胞损伤模型;利用四甲基偶氮唑蓝(MTT)法考察预孵育二氢槲皮素对H2O2诱导H9c2心肌细胞损伤细胞存活率的影响;利用试剂盒检测培养液中乳酸脱氢酶( LDH),细胞内丙二醛( MDA),过氧化氢酶( CAT )、超氧化物歧化酶( SOD )及谷胱甘肽过氧化物酶( GSH-Px )含量的变化。结果:不同浓度的二氢槲皮素预处理4、12、24 h均能够提高细胞的存活率,且呈剂量依赖性,其中预处理12 h的保护作用最为明显。与正常对照组相比,模型组的存活率下降,LDH的漏出量增大,细胞内MDA的含量增大,CAT、SOD、GSH-Px的活力明显降低。用二氢槲皮素预处理12 h能显著地减少LDH的漏出量,降低细胞内MDA的含量,提升细胞内CAT、SOD、GSH-Px的活力,并呈剂量依赖性。结论:二氢槲皮素对H2 O2诱导H9 c2心肌细胞损伤具有显著保护作用,且呈剂量依赖性,其作用机制可能与其能够提高细胞内抗氧化酶活性,增强细胞稳定性有关。

목적:연구이경곡피소대H2 O2유도H9c2심기세포손상적보호작용。방법:건립H2 O2유도H9c2심기세포손상모형;이용사갑기우담서람(MTT)법고찰예부육이경곡피소대H2O2유도H9c2심기세포손상세포존활솔적영향;이용시제합검측배양액중유산탈경매( LDH),세포내병이철( MDA),과양화경매( CAT )、초양화물기화매( SOD )급곡광감태과양화물매( GSH-Px )함량적변화。결과:불동농도적이경곡피소예처리4、12、24 h균능구제고세포적존활솔,차정제량의뢰성,기중예처리12 h적보호작용최위명현。여정상대조조상비,모형조적존활솔하강,LDH적루출량증대,세포내MDA적함량증대,CAT、SOD、GSH-Px적활력명현강저。용이경곡피소예처리12 h능현저지감소LDH적루출량,강저세포내MDA적함량,제승세포내CAT、SOD、GSH-Px적활력,병정제량의뢰성。결론:이경곡피소대H2 O2유도H9 c2심기세포손상구유현저보호작용,차정제량의뢰성,기작용궤제가능여기능구제고세포내항양화매활성,증강세포은정성유관。
Objective:To investigate the protective effects of dihydroquercetin against H 2 O2 -induced injury in H9c2 cardiomyoblasts.Methods:A model of H2O2 -induced injury in H9c2 cardiomyoblasts was established. Cell viability was determined colorimetrically by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazo-lium (MTT) assay.The supernates and cells were collected , respectively, after the different treatments for measuring the LDH and MDA levels, as well as SOD,CAT, and GSH-Px activity with the corresponding de-tection kit according to the manufacturer ’ s instructions .Results:H9 c2 cells were pretreated with dihydroquer-cetin for 12 h before exposure to 150 μM for 6 h.H2 O2 treatment caused severe injury to H9c2 cardiomyo-blasts.Dihydroquercetin pretreatment could attenuate H 2 O2 -induced injury , decrease the levels of LDH leakage and MDA, and increase the activities of SOD, CAT, and GSH-Px with a dose-dependent.Conclu-sion:Dihydroquercetin showed significantly protective effects against H 2 O2 -induced injury in H9c2 cardio-myoblasts in dose-dependent .The mechanism of this may be due to its increasing the activities of endogenous antioxidant enzymes , as well as improving the stabilization of myocardial cells .
