目的:探讨丹参酮ⅡA对体外培养星形胶质细胞细胞周期和增殖的影响。方法本研究采用体外细胞培养方法获得新生SD小鼠大脑皮质星形胶质细胞,然后在培养液中分别加入不同浓度的丹参酮ⅡA,作用8 h及24 h后,通过流式细胞仪检测丹参酮ⅡA对星形胶质细胞细胞周期的影响。结果丹参酮ⅡA作用于星形胶质细胞后,可引起细胞周期的显著变化,表现为G0/G1期细胞数减少,S期、G2/M期细胞数增多。结论丹参酮ⅡA可促进体外培养的星形胶质细胞增殖。

목적:탐토단삼동ⅡA대체외배양성형효질세포세포주기화증식적영향。방법본연구채용체외세포배양방법획득신생SD소서대뇌피질성형효질세포,연후재배양액중분별가입불동농도적단삼동ⅡA,작용8 h급24 h후,통과류식세포의검측단삼동ⅡA대성형효질세포세포주기적영향。결과단삼동ⅡA작용우성형효질세포후,가인기세포주기적현저변화,표현위G0/G1기세포수감소,S기、G2/M기세포수증다。결론단삼동ⅡA가촉진체외배양적성형효질세포증식。
Aim To study the effects of tanshinoneⅡA on cell cycles and proliferation of astrocyte in vitro. Methods Cerebral cortex astrocytes of neonatal rats were generated through cell culture in this study. Different concentrations of tanshinoneⅡA were administrated to cultured astrocytes. The effects of tanshinoneⅡA on the cell cycle of astrocytes were detected by means of lfow cytometer at 8 h or 24 h following drug application. Results The results showed that significant changes of cell cycle occurred following administering tanshinoneⅡA. The percentage of cells in G0/G1 cycle phase was reduced and the percentage of S and G2/M cycle phase increased. Conclusion TanshinoneⅡA may promote the proliferation of cultured cerebral cortex astrocytes.
