以玉米C型细胞质雄性不育系及保持系为材料,利用实时荧光定量聚合酶链反应检测 pcd5,cox1和mdh基因在小孢子单核期的表达水平。结果表明,相对于保持系,这3个基因在不育系中均下调表达。利用焦磷酸测序与高分辨率溶解曲线相结合的方法证明了在不育系单核期 pcd5转录本GRMZM2G075839 T02的5′非编码区域存在A碱基插入突变。该突变可能与 pcd5基因在不育系中下调表达有关。这些发现可为进一步研究细胞程序性死亡与玉米C型胞质花粉败育的关系提供参考。

Summary Programmed cell death ( PCD) is crucial for plant growth and development . It maintains plant normal physiologic metabolism by eliminating damaged , diseased and old cells . Numerous studies have demonstrated that cytoplasmic male sterility (CMS) in plant was associated with programmed cell death ( PCD) . The proper timing of PCD is very important for floral development .Based on RNA-Seq (RNA sequencing) result , we found that programmed cell death 5 gene pcd5 , cytochrome oxidase gene cox1 and malate dehydrogenase gene mdh were all down-regulated in CMS-C line of maize . In this study , real-time fluorescence quantitative polymerase chain reaction ( RT-qPCR) was done to further validate the expression of pcd5 ,cox1 and mdh genes . These analyses will contribute to better understand CMS-C mechanisms .
