为建立一种环境友好型防治黄颡鱼!爱德华氏菌病的方法,利用从患病黄颡鱼脑部组织分离出的致病菌株JXHS制备黄颡鱼!爱德华氏菌的灭活疫苗。以平均体重为15~20 g的健康黄颡鱼为实验对象,免疫组腹腔注射0.2 mL浓度为3.0×109 CFU·mL-1的免疫原,对照组注射等量0.65%灭菌生理盐水,分别在注射7、14、21、28 d后随机从两组各取30尾实验鱼,尾静脉采血,检测免疫鱼的血清凝集抗体效价、白细胞吞噬活性(PP)、吞噬指数(PI)、溶菌酶(LSZ)、碱性磷酸酶(AKP)、酸性磷酸酶(ACP)等免疫指标的变化。结果表明:!爱德华氏菌灭活疫苗对黄颡鱼具有较强的免疫原性,与对照组相比,血清凝集抗体效价在免疫14、21和28 d显著升高;PP、PI和LSZ在所有采样时间点均显著上升;LSZ在免疫7、14、21和28 d均显著上升;AKP在免疫14、21和28 d活力显著上升;ACP在免疫7、14和21 d活力显著上升。血清凝集抗体效价、PP、PI、LSZ和 AKP均在21 d达到峰值,而 ACP在免疫7 d后达到峰值。免疫30 d后!爱德华氏菌攻毒结果表明,!爱德华氏菌灭活疫苗对黄颡鱼有较高的免疫保护作用,免疫保护率达到70.2%。由此可见,利用从病鱼体内分离的!爱德华氏菌菌株制备的灭活疫苗可提高黄颡鱼的免疫力并有效抵御!爱德华氏菌的攻击。

위건립일충배경우호형방치황상어!애덕화씨균병적방법,이용종환병황상어뇌부조직분리출적치병균주JXHS제비황상어!애덕화씨균적멸활역묘。이평균체중위15~20 g적건강황상어위실험대상,면역조복강주사0.2 mL농도위3.0×109 CFU·mL-1적면역원,대조조주사등량0.65%멸균생리염수,분별재주사7、14、21、28 d후수궤종량조각취30미실험어,미정맥채혈,검측면역어적혈청응집항체효개、백세포탄서활성(PP)、탄서지수(PI)、용균매(LSZ)、감성린산매(AKP)、산성린산매(ACP)등면역지표적변화。결과표명:!애덕화씨균멸활역묘대황상어구유교강적면역원성,여대조조상비,혈청응집항체효개재면역14、21화28 d현저승고;PP、PI화LSZ재소유채양시간점균현저상승;LSZ재면역7、14、21화28 d균현저상승;AKP재면역14、21화28 d활력현저상승;ACP재면역7、14화21 d활력현저상승。혈청응집항체효개、PP、PI、LSZ화 AKP균재21 d체도봉치,이 ACP재면역7 d후체도봉치。면역30 d후!애덕화씨균공독결과표명,!애덕화씨균멸활역묘대황상어유교고적면역보호작용,면역보호솔체도70.2%。유차가견,이용종병어체내분리적!애덕화씨균균주제비적멸활역묘가제고황상어적면역력병유효저어!애덕화씨균적공격。
The research aimed to establish an environment-friendly way to protect the diseases of yellow cat-fish (Pelteobagrus fulvidraco)with Edwardsiella ictaluri.Healthy yellow catfish were intraperitoneally injected with 0.2ml (3.0×109 CFU·mL-1)inactivated vaccine of Edwardsiellaictaluri JXHS strain, which was isolated from yellow catfish with"Cracked head disease".The control group was inj ected with same volume of 0.65% physiological saline.Samples were collected after vaccination 7,14,21 and 28 days. Serum antibody,blood leucocytes phagocytosis,serum lysozyme activity,alkaline phosphatase activity and acid phosphatase activity were assayed respectively.The results showed that the serum antibody titers were significantly increased at 14,21 and 28d;phagocytic percentage,phagocytic index and lysozyme activity were significantly enhanced at all the time points,peaking at 2 1 day;alkaline phosphatase activity signifi-cantly increased at 14,21 and 28d;and acid phosphatase activity was higher than the control group’s at 7, 14 and 21d.At 30 day post-immunization,healthy yellow catfish challenged with Edwardsiellaictaluri to detect the protective effects of the vaccine.It was found that the immune protection rate of immunized group was 70.2%.In conclusion,Edwardsiella ictaluri inactivated vaccine can effectively prevent yellow catfish from Edwardsiellosis.
