目的:通过四种建模方法的比较,明确建立颞下颌关节滑膜炎动物模型的方法,并探讨白介素1β(IL-1β)在滑膜炎发病机制中的作用。方法:将25只8周龄的雄性Wistar大鼠随机分为5组,即对照组、咬肌切除组、咬合升高组、咬肌切除加咬合升高组、咬合创伤组。大鼠12周龄时取双侧关节, HE染色比较各组组织病理学变化、病理分值间的差异,免疫组化比较各组IL-1β表达。结果:咬肌切除加咬合升高组病理分值、IL-1β平均光密度值与其他各组比较差异均有统计学意义(P<0.05),各组病理分值与IL-1β平均光密度值之间差异有统计学意义(r=0.80, P<0.001)。结论:咬肌切除加咬合升高可造成滑膜炎,此方法可作为建立滑膜炎的有效方法, IL-1β在滑膜炎发病中起到了关键作用。

목적:통과사충건모방법적비교,명학건립섭하합관절활막염동물모형적방법,병탐토백개소1β(IL-1β)재활막염발병궤제중적작용。방법:장25지8주령적웅성Wistar대서수궤분위5조,즉대조조、교기절제조、교합승고조、교기절제가교합승고조、교합창상조。대서12주령시취쌍측관절, HE염색비교각조조직병이학변화、병리분치간적차이,면역조화비교각조IL-1β표체。결과:교기절제가교합승고조병리분치、IL-1β평균광밀도치여기타각조비교차이균유통계학의의(P<0.05),각조병리분치여IL-1β평균광밀도치지간차이유통계학의의(r=0.80, P<0.001)。결론:교기절제가교합승고가조성활막염,차방법가작위건립활막염적유효방법, IL-1β재활막염발병중기도료관건작용。
Objective:to compare four methods that are to establish a animal model of temporomandibular joint synovitis, and to explore the role of IL-1βin pathogenesis of synovitis.Methods:25 male 8-week-old Wistar rats were randomly divided into 5 groups: control group, masseter muscle resection group, occlusal vertical dimension increase group, masseter resection plus increased occlusal vertical dimension group, and occlusal interference group. HE staining were performed on the specimens from 12-week-old mice tissues. The pathological changes, pathological score differences between groups, and the immunohistochemical expression of IL-1βwere compared among the groups.Results:Masseter resection plus vertical dimension increase group were statistically different than the other groups in pathological score and the average optical density of IL-1β(P<0.05). There was a significantly positive correlation between pathological score and the average optical density value of IL-1β(r=0.80,P<0.001).Conclusions:The masseter resection plus the occlusal vertical dimension increase can cause synovitis. This method can be used as an effective method to establish TMJ synovitis, and IL-1βplays a key role in TMJ synovitis.
