目的:观察愈结灵对实验性结肠炎(UC)小鼠的治疗作用及机制。方法:C57BL/C小鼠40只随机分为正常、模型、SASP(520 mg/kg),YJL组,每组10只,除正常组外,余以5%DSS水溶液自由饮用8 d产生UC模型,同时灌服相应药物。评价各组小鼠的体质量变化,观察结肠组织病理学改变,疾病活动指数评分(disease activity index, DAI),Western blotting法检测结肠组织中核转录因子(NF-κB)的表达。结果:愈结灵能显著减轻结肠炎小鼠结肠组织病理学损伤。与模型组相比,各给药组DAI明显降低( p<0.01),结肠组织NF-κB表达下降( p<0.01)。结论:TASA可能通过抑制NF-κB的表达,发挥治疗溃疡性结肠炎的作用。

목적:관찰유결령대실험성결장염(UC)소서적치료작용급궤제。방법:C57BL/C소서40지수궤분위정상、모형、SASP(520 mg/kg),YJL조,매조10지,제정상조외,여이5%DSS수용액자유음용8 d산생UC모형,동시관복상응약물。평개각조소서적체질량변화,관찰결장조직병이학개변,질병활동지수평분(disease activity index, DAI),Western blotting법검측결장조직중핵전록인자(NF-κB)적표체。결과:유결령능현저감경결장염소서결장조직병이학손상。여모형조상비,각급약조DAI명현강저( p<0.01),결장조직NF-κB표체하강( p<0.01)。결론:TASA가능통과억제NF-κB적표체,발휘치료궤양성결장염적작용。
Objective:To investigate the therapeutic effects of Yu Jie Ling on mice with ulcerative colitis. Methods:Forty male C57BL/C mice were randomly divided into four groups: normal control group, UC model group, Sulfasalazine (SASP) group and Yu Jie Ling (YJL) group. The normal control was drinking distilled water freely, while the other groups were administrated 5%dextran sulfate sodium(DSS)solution as drinking water freely for 8 days to induce acute ulcerative colitis. At the same time the mice were given oral YJL (in the model or control groups), SASP in SASP group. The general morphological and histopathological changes were observed during the experiment. Daily disease activity index (DAI) was evaluated. Nuclear factor (NF-κB) of colonic expression was detected by Western blotting. Results:The histopathological changes in YJL group had great amelioration, compared with model group. The DAI decreased significantly, and the expression of NF-κB in colon was all obviously reduced. Conclusion:Yu Jie Ling can effectively attenuate DSS-induced ulcerative colitis through the decrease of the expression of NF-κB.
