目的:观察地锦草水提液对糖尿病小鼠的降血糖作用,并探究其作用机制。方法对60只小鼠进行尾静脉注射0.25%四氧嘧啶溶液80 mg·kg-1,注射后2 h给予50%葡萄糖溶液0.2 mL·10 g-1灌胃造模,3 d后眼眶采血测血糖,血糖值>11.1 mmol·L-1为糖尿病小鼠。将50只造模成功的小鼠按随机数字表法分为5组:NS组(阴性对照组),地锦草低(59 mg·kg-1·d-1)、中(118 mg·kg-1·d-1)、高(236 mg·kg-1·d-1)剂量组,二甲双胍组(阳性对照组),每组10只。连续给药14 d后,观察地锦草水提液对糖尿病小鼠血糖、血清胰岛素、胰岛病理形态变化的影响。结果1)备糖:地锦草高剂量组和二甲双胍组给药后的血糖明显低于给药前及 NS 组,地锦草低、中剂量组(均P<0.05),NS组、地锦草中、低剂量组血糖也略下降,但差异均无统计学意义(均P>0.05)。2)血清胰岛素:地锦草高剂量组、二甲双胍组血清胰岛素水平均显著高于 NS 组及地锦草低、中剂量组(均P<0.05),地锦草低、中剂量组及 NS组比较差异均无统计学意义(均P>0.05)。3)胰腺的病理形态:NS组胰岛数量明显减少,结构破坏,边界模糊不清,胰岛β细胞排列紊乱,部分细胞出现变性或坏死;地锦草各剂量组胰岛数量较多,胰岛结构较完整,境界较清晰,胰岛β细胞排列较整齐,细胞形态较好,胞浆丰富,胞核多数清楚,仅可见少数细胞变性或坏死;二甲双胍组胰岛形态有一定的恢复。结论地锦草水提液对糖尿病小鼠的降血糖作用可能是通过保护胰岛β细胞、减轻其损伤及增加胰岛素分泌来实现。

목적:관찰지금초수제액대당뇨병소서적강혈당작용,병탐구기작용궤제。방법대60지소서진행미정맥주사0.25%사양밀정용액80 mg·kg-1,주사후2 h급여50%포도당용액0.2 mL·10 g-1관위조모,3 d후안광채혈측혈당,혈당치>11.1 mmol·L-1위당뇨병소서。장50지조모성공적소서안수궤수자표법분위5조:NS조(음성대조조),지금초저(59 mg·kg-1·d-1)、중(118 mg·kg-1·d-1)、고(236 mg·kg-1·d-1)제량조,이갑쌍고조(양성대조조),매조10지。련속급약14 d후,관찰지금초수제액대당뇨병소서혈당、혈청이도소、이도병리형태변화적영향。결과1)비당:지금초고제량조화이갑쌍고조급약후적혈당명현저우급약전급 NS 조,지금초저、중제량조(균P<0.05),NS조、지금초중、저제량조혈당야략하강,단차이균무통계학의의(균P>0.05)。2)혈청이도소:지금초고제량조、이갑쌍고조혈청이도소수평균현저고우 NS 조급지금초저、중제량조(균P<0.05),지금초저、중제량조급 NS조비교차이균무통계학의의(균P>0.05)。3)이선적병리형태:NS조이도수량명현감소,결구파배,변계모호불청,이도β세포배렬문란,부분세포출현변성혹배사;지금초각제량조이도수량교다,이도결구교완정,경계교청석,이도β세포배렬교정제,세포형태교호,포장봉부,포핵다수청초,부가견소수세포변성혹배사;이갑쌍고조이도형태유일정적회복。결론지금초수제액대당뇨병소서적강혈당작용가능시통과보호이도β세포、감경기손상급증가이도소분비래실현。
Objective To explore the hypoglycemic effect of humifuse euphorbia herb extract in diabetic mice and its mechanism of action.Methods Sixty mice were given intravenous inj ection of 0.25% alloxan solution (80 mg·kg-1 )though tail vein,and then were intragastricly adminis-trated 50% glucose solution(0.2 mL·10 g-1)2 hours after intravenous injection.After 3 days, orbital blood was taken to detect the levels of blood glucose.Mice with blood glucose levels above 11.1 mmol·L-1 were regarded as the diabetic mice.Fifty diabetic mice were randomly divided in-to 5 groups:normal saline group (negative control group ),low-dose humifuse euphorbia herb group (59 mg·kg-1·d-1),medium-dose humifuse euphorbia herb group(118 mg·kg-1·d-1), high-dose humifuse euphorbia herb group(236 mg·kg-1 ·d-1)and metformin group(positive control group),with 10 mice in each group.After successive administration for 14 days,levels of blood glucose and serum insulin were measured and pathological changes in pancreatic islet wereobserved.Results 1)Blood glucose:Compared with both negative control group and low,medi-un-dose humifuse euphorbia herb group and before medication,blood glucose levels significantly decreased in both high-dose humifuse euphorbia herb group and metformin group (P<0.05 ). However,no obvious decrease in blood glucose levels were found after treatment in negative con-trol group,low-dose humifuse euphorbia herb group and medium-dose humifuse euphorbia herb group(all P>0.05).2)insulin:Compared with negative control group,low-dose humifuse eu-phorbia herb group or medium-dose humifuse euphorbia herb group,serum insulin levels marked-ly increased in both high-dose humifuse euphorbia herb group and metformin group (all P<0.05 ).No significant differences in serum insulin levels were found among negative control group,low-dose humifuse euphorbia herb group and medium-dose humifuse euphorbia herb group (all P>0.05).3)Pathological change of the pancreas:Pathological analysis showed obvious de-crease in the number the pancreatic islets with damaged structures,blurred boundaries,disorderedβcell arrangement and partial degeneration or necrosis in negative control group,but significant increase in the number the pancreatic islets with complete structures,clear boundaries,orderlyβcell arrangement,normal cell morphology,abundant cytoplasm,clear nucleus and mild degenera-tion or necrosis in humifuse euphorbia herb groups.In addition,metformin treatment improved the morphology of pancreatic islets compared with negative control group.Conclusion Humifuse euphorbia herb extract exerts has hypoglycemic effect in diabetic mice through protecting pancre-atic isletβcells,reducing damage and promoting insulin secretion.
