Cymbidium f aberi%TUA gene%cloning%sequence analysis
α微管蛋白(TUA)基因常作为研究其他功能基因表达的内参基因。以TUA作为候选内参基因之一,依据GenBank已经公布的同源TUA基因的核苷酸保守序列,设计简并性引物,以蕙兰花蕾期的花葶为材料,提取总RNA,利用RTPCR的方法,克隆了4条TUA基因片段。序列分析结果表明,4条TUA基因片段长度均为1205bp,编码401个氨基酸,具有TUA基因标记位点:Tubulinsubunitsalpha,beta,andgammasignature特征信号序列(GGGTGSG)。各序列之间同源性高达99.63%,经Blast分析,与水稻同源性可达86%。利用DNAMAN等生物软件推断的氨基酸,与月季、玉米、牛筋草、小麦、看麦娘和水稻TUA基因所编码的氨基酸序列同源性达98%。研究中所得到的4个基因序列是TUA基因的同源片段,依次命名为CfTUA1、CfTUA2、CfTUA3、CfTUA4,GenBank登录号分别为JN177709、JN177710、JN177711、JN177712。

TUA is often used as an internal reference gene to analyze expression level of the functional genes .In this paper ,RNA was extracted from the pedicels of Cymbidium f aberi ,and four TUA genes were cloned based on homologous cloning with a pair of primers designed on the basis of the conserved segments of the TUA genes in NCBI webset .The sequence analysis result revealed that the length of four TUA genes was all 1 205 bp ,which encods a protein of 401 amino acids .Four TUA genes all contained tubulin subunits alpha ,beta ,and gamma signature(GGGTGSG) . The nucleotide sequence of four TUA genes had 99.63% identity to each other and 86% identity to that from Oryza sativa .The deduced amino acid sequence of four TUA genes had 98% identity to that from Rosa hybrid ,Zea mays ,Eleusine indica ,Triticum aestivum ,Alopecurus aequalis and Oryza sativa .The cloned genes were TUA gene fragments ,named as Cf TUA1 ,Cf TUA2 ,Cf TUA3 and Cf TUA5 respectively . They were registered into GenBank (respective accession number :JN177709 ,JN177710 ,JN177711 and JN177712) .
