目的:观察丁苯酞( NBP)对局灶性脑缺血再灌注损伤大鼠脑组织Fas和FasL表达的影响。方法30只SD大鼠随机分为假手术组( Sham组)、脑缺血再灌注组( I/R组)和丁苯酞治疗组( NBP组)。线栓法制作大鼠大脑中动脉脑缺血再灌注模型,于缺血2 h再灌注24 h后断头取脑,免疫组织化学法检测缺血区脑组织Fas和FasL的蛋白表达,逆转录聚合酶链反应( RT-PCR)法检测缺血区脑组织Fas和FasL的mRNA表达。结果与Sham组相比, I/R组缺血区脑组织Fas和FasL的蛋白和mRNA的表达均显著增多;与I/R组相比,NBP组缺血区脑组织Fas和FasL的蛋白和mRNA的表达均显著下调,差异均有统计学意义( P<0.05)。结论丁苯酞对大鼠脑缺血再灌注损伤的保护作用,其机制可能与抑制Fas和FasL的表达有关。

목적:관찰정분태( NBP)대국조성뇌결혈재관주손상대서뇌조직Fas화FasL표체적영향。방법30지SD대서수궤분위가수술조( Sham조)、뇌결혈재관주조( I/R조)화정분태치료조( NBP조)。선전법제작대서대뇌중동맥뇌결혈재관주모형,우결혈2 h재관주24 h후단두취뇌,면역조직화학법검측결혈구뇌조직Fas화FasL적단백표체,역전록취합매련반응( RT-PCR)법검측결혈구뇌조직Fas화FasL적mRNA표체。결과여Sham조상비, I/R조결혈구뇌조직Fas화FasL적단백화mRNA적표체균현저증다;여I/R조상비,NBP조결혈구뇌조직Fas화FasL적단백화mRNA적표체균현저하조,차이균유통계학의의( P<0.05)。결론정분태대대서뇌결혈재관주손상적보호작용,기궤제가능여억제Fas화FasL적표체유관。
Objective To investigate the effects of dl -3 -n -bu-tylphthalide on the expression of Fas and FasL in rats with focal cerebral ischemia -reperfusion.Methods Thirty rats were randomly assigned into three groups:Sham operation group (Sham group),focal cerebral is-chemia-reperfusion group ( I/R group ) and dl-3-n-butylphthalide group ( NBP group ).The middle cerebral artery occlusion reperfusion model was made by the suture method (ischemia for 2 hours, and reper-fusion for 24 hours ).The level of Fas and FasL expression were meas-ured by immunohistochemistry and reverse transcription -polymerase chain reaction(RT-PCR).Results Compared with sham group, the expressions of Fas and FasL as well as the mRNA were significantly up -regulated in I/R group ( P<0.05 ).Compared with I/R group, the ex-pressions of Fas and FasL as well as the mRNA were significantly down -regulated in the NBP group ( P<0.05 ).Conclusion The protection effect of dl-3-n-butylphthalide on rats with focal cerebral ischemia -reperfusion may be related to the inhibition of the expression of Fas and FasL.
