目的:研究临床分离的阴沟肠杆菌 AcrAB-TolC 外排系统基因的携带情况及其与抗菌药物耐药性的关系。方法对临床分离的141株阴沟肠杆菌用 MicroScanWalkAway40全自动细菌分析仪进行鉴定和药敏试验;聚合酶链式反应(PCR)检测 AcrAB-TolC 外排系统 acrA,acrB,tolC 和 acrR 4种基因在阴沟肠杆菌中的分布,并对 PCR 产物进行纯化、克隆、测序。结果 acrA,acrB,tolC 和 acrR 4种基因在141株阴沟肠杆菌中的检出率分别为98.5%、51.06%、82.27%、78.72%,且4种基因检出率有显著性差异(P <0.01);acrA 基因和 acrB 基因在不同组的阳性率有显著性差异(P <0.01),各耐药组高于敏感组,且多重耐药组阳性率最高;acrR 基因和 tolC 基因在不同组的阳性率无显著性差异。结论我院分离的阴沟肠杆菌高水平携带 AcrAB-TolC 外排系统基因,且与抗菌药物耐药性具有相关性。
目的:研究臨床分離的陰溝腸桿菌 AcrAB-TolC 外排繫統基因的攜帶情況及其與抗菌藥物耐藥性的關繫。方法對臨床分離的141株陰溝腸桿菌用 MicroScanWalkAway40全自動細菌分析儀進行鑒定和藥敏試驗;聚閤酶鏈式反應(PCR)檢測 AcrAB-TolC 外排繫統 acrA,acrB,tolC 和 acrR 4種基因在陰溝腸桿菌中的分佈,併對 PCR 產物進行純化、剋隆、測序。結果 acrA,acrB,tolC 和 acrR 4種基因在141株陰溝腸桿菌中的檢齣率分彆為98.5%、51.06%、82.27%、78.72%,且4種基因檢齣率有顯著性差異(P <0.01);acrA 基因和 acrB 基因在不同組的暘性率有顯著性差異(P <0.01),各耐藥組高于敏感組,且多重耐藥組暘性率最高;acrR 基因和 tolC 基因在不同組的暘性率無顯著性差異。結論我院分離的陰溝腸桿菌高水平攜帶 AcrAB-TolC 外排繫統基因,且與抗菌藥物耐藥性具有相關性。
목적:연구림상분리적음구장간균 AcrAB-TolC 외배계통기인적휴대정황급기여항균약물내약성적관계。방법대림상분리적141주음구장간균용 MicroScanWalkAway40전자동세균분석의진행감정화약민시험;취합매련식반응(PCR)검측 AcrAB-TolC 외배계통 acrA,acrB,tolC 화 acrR 4충기인재음구장간균중적분포,병대 PCR 산물진행순화、극륭、측서。결과 acrA,acrB,tolC 화 acrR 4충기인재141주음구장간균중적검출솔분별위98.5%、51.06%、82.27%、78.72%,차4충기인검출솔유현저성차이(P <0.01);acrA 기인화 acrB 기인재불동조적양성솔유현저성차이(P <0.01),각내약조고우민감조,차다중내약조양성솔최고;acrR 기인화 tolC 기인재불동조적양성솔무현저성차이。결론아원분리적음구장간균고수평휴대 AcrAB-TolC 외배계통기인,차여항균약물내약성구유상관성。
Objective To study the carrying situation of AcrAB-TolC efflux system genes in Enterobacter cloacae of the clinical isolates,and investigate the relationship between the genes and the drug resistance of E.cloacae.Methods The141 clinical isolates of E.cloacae were given the identification and antibiotic susceptibility testing by MicroScan WalkAway 40 system.PCR was used to detected acrA,acrB,tolC and acrR gene in the isolates,and the product of PCR was confirmed by sequenced after purifying and cloning.Results The detection rates of acrA,acrB,tolC and acrR genes of the141 clinical isolates were 98.5%、51.06%、82.27%、78.72% respectively.And the detection rate of four genes had significant differences(P <0.01).The detection rates of drug groups were higher than the sensitive group.And the detection rate of the multidrug resistant group is the highest.However,there were no significant differences in the de-tection rates of acrR and tolC genes in different groups.Conclusion The E.cloacae stains which isolated from our hos-pital carried a high level of AcrAB-TolC efflux system genes,and it was related with the antimicrobial drug resistance.
