As a kind of malignant tumor derived from mesenchymal tissues, osteosarcoma is harmful to adolescent health and may cause death, with a higher grade of malignancy and metastasis and worse treatment outcomes and prognosis. Chemotherapy, especially neoadjuvant chemotherapy, plays a very important role in the treatment of osteosarcoma. However, because of the deifciency of treatment speciifcity, the antineoplastic drug will do harm to normal cells and organs and meanwhile kill osteosarcoma cells. Depending on selective functions of the oriented part, targeted therapies will selectively act on target locations and toxic reactions will also be reduced for normal organs. Different mechanisms of targeted therapies will be used on different target points. The key examples of target identiifcation in osteosarcomas is highlighted in this review. The drug doses and side effects can be reduced and the treatment effects can be improved, with potential clinical values in the future.
