目的:观察双氢青蒿素(dihydroartemisinin ,DHA)对C2株蓝氏贾第鞭毛虫(Giardia lamblia)Alpha-7.3 giar-din (α-贾第素)基因mRNA表达水平的影响,探讨其对蓝氏贾第鞭毛虫骨架蛋白的损伤作用。方法用双氢青蒿素浓度为100μg/mL、200μg/mL的改良TYI-S-33培养基分别培养C2株蓝氏贾第鞭毛虫2 h、4 h、8 h、12 h后,以不含药物组为对照,实时荧光定量RT-PCR检测药物作用后Alpha-7.3 giardin基因mRNA表达水平的变化。结果双氢青蒿素作用虫体后Al-pha-7.3 giardin基因mRNA表达水平明显低于对照组,二者有显著性差异。结论双氢青蒿素对C2株蓝氏贾第鞭毛虫Al-pha-7.3 giardin基因mRNA的表达具有明显的抑制作用,抑制效果与药物浓度和作用时间相关,提示双氢青蒿素对蓝氏贾第鞭毛虫骨架蛋白具有损伤作用。
目的:觀察雙氫青蒿素(dihydroartemisinin ,DHA)對C2株藍氏賈第鞭毛蟲(Giardia lamblia)Alpha-7.3 giar-din (α-賈第素)基因mRNA錶達水平的影響,探討其對藍氏賈第鞭毛蟲骨架蛋白的損傷作用。方法用雙氫青蒿素濃度為100μg/mL、200μg/mL的改良TYI-S-33培養基分彆培養C2株藍氏賈第鞭毛蟲2 h、4 h、8 h、12 h後,以不含藥物組為對照,實時熒光定量RT-PCR檢測藥物作用後Alpha-7.3 giardin基因mRNA錶達水平的變化。結果雙氫青蒿素作用蟲體後Al-pha-7.3 giardin基因mRNA錶達水平明顯低于對照組,二者有顯著性差異。結論雙氫青蒿素對C2株藍氏賈第鞭毛蟲Al-pha-7.3 giardin基因mRNA的錶達具有明顯的抑製作用,抑製效果與藥物濃度和作用時間相關,提示雙氫青蒿素對藍氏賈第鞭毛蟲骨架蛋白具有損傷作用。
목적:관찰쌍경청호소(dihydroartemisinin ,DHA)대C2주람씨가제편모충(Giardia lamblia)Alpha-7.3 giar-din (α-가제소)기인mRNA표체수평적영향,탐토기대람씨가제편모충골가단백적손상작용。방법용쌍경청호소농도위100μg/mL、200μg/mL적개량TYI-S-33배양기분별배양C2주람씨가제편모충2 h、4 h、8 h、12 h후,이불함약물조위대조,실시형광정량RT-PCR검측약물작용후Alpha-7.3 giardin기인mRNA표체수평적변화。결과쌍경청호소작용충체후Al-pha-7.3 giardin기인mRNA표체수평명현저우대조조,이자유현저성차이。결론쌍경청호소대C2주람씨가제편모충Al-pha-7.3 giardin기인mRNA적표체구유명현적억제작용,억제효과여약물농도화작용시간상관,제시쌍경청호소대람씨가제편모충골가단백구유손상작용。
Effects of dihydroartemisinin (DHA) on the expression level of Alpha-7 .3 giardin mRNA in C2 Giardia lam-blia was investigated in this study to explore the damage to skeleton protein of C 2 Giardia lamblia .Giardia lamblia was culti-vated respectively for 2 ,4 ,8 ,and 12 hours with modified TYI-S-33 medium containing 100 μg/mL and 200 μg/mL DHA , while the control group performed in the same experimental conditions without DHA .The expressive quantity of Alpha-7 .3 gi-ardin mRNA was determined by using real-time reverse transcription PCR ,and then we found that the expressive quantities of Alpha-7 .3 giardin mRNA with DHA were significantly lower than those in the control group .It’s suggested that dihydroarte-misinin has obvious inhibitory effect on the expression level of Alpha-7 .3 giardin mRNA in C2 Giardia lamblia .The actions of dihydroartemisinin on skeleton protein of C2 Giardia lamblia are effective .
