目的:了解 IL-28B 基因中5个基因位点的连锁不平衡与 HCV 感染的相关性。方法利用聚合酶链扩增和测序方法分别测定88例 HCV 感染者和145例健康对照人群的 IL-28B 基因中 rs8099917、rs8113007、rs12972991、rs12979860和 rs12980275五个位点的基因型,通过连锁不平衡分析软件分析 HCV 感染组和正常对照组中5个 IL-28B 基因位点的连锁不平衡状态和单倍体型频率。结果 HCV 感染组和正常对照组中的单倍体型分布频率差异有统计学意义(P <0.01)。HCV 感染的患者和正常组中 IL-28B 基因5个位点间存在基因连锁不平衡,其中 rs12979860与其他4个位点之间连锁不平衡程度在正常人群组和 HCV 感染组中差异有统计学意义(P <0.01);rs8113007与其他4个位点之间的连锁不平衡程度在正常组和 HCV 感染组中差异有统计学意义(P <0.05)。结论相对于在 HCV 感染者,IL-28B 基因中rs12979860位点和 rs8113007位点与其他位点的连锁不平衡在健康人群中可能是更好的保护性模式。

Objective To study the correlation of IL-28B gene loci linkage disequilibrium and HCV infection.Methods The IL-28B rs8099917,rs8113007,rs12972991 ,rs12979860 and rs12980275 loci genotype in 88 HCV infected patients and 145 healthy volunteers were determined by sequencing method,Linkage disequilibrium of IL-28B gene loci and haploid frequency in HCV infection group and healthy group were analyzed using a linkage disequilibrium analysis software.Results 1 .There was a significant difference in haplotype frequency between HCV infection group and healthy group (P <0.01). 2.There was linkage disequilibrium of IL-28B gene in patients with HCV and healthy group between rs12979860 and other loci including rs8099917,rs12980275,rs8113007 and rs12972991 .Linkage disequilibrium between rs12979860 and other four loci in the healthy volunteers was significantly different (P <0.01)than that in HCV infection group.The difference of disequilibrium degree between rs8113007 and the other four loci was significant between the healthy group and HCV infection group (P < 0.05 ).Conclusion With respect to the HCV infection,linkage disequilibrium of IL-28B gene rs12979860 locus and rs8113007 locus in healthy volunteers may be a better protecting mode.
