目的：探讨细胞周期蛋白 G2(cyclin G2,CCNG2)在结直肠癌中的表达意义。方法采用免疫组织化学方法、蛋白印迹法(Western blot)检测57例结直肠癌组织及距其癌组织边缘2 cm 以上的镜下未见癌浸润的正常组织中蛋白的表达情况。结果免疫组织化学结果表明,CCNG2蛋白在结直肠癌组织中的表达较正常结直肠组织明显降低,分别为38.6%(22/57)、91.3%(31/33)(P <0.05)。Western blot 结果表明,CCNG2的蛋白在结直肠癌组织的表达相对量较正常结直肠组织的表达相对量明显降低(P <0.05)。CCNG2蛋白表达与结直肠癌 T 分期、淋巴结转移、肿瘤临床分期及肿瘤分化有关(P <0.05)。结论结直肠癌组织中 CCNG2蛋白表达明显减低,且与结直肠癌 T 分期、淋巴结转移、临床分期及肿瘤分化有关。
목적：탐토세포주기단백 G2(cyclin G2,CCNG2)재결직장암중적표체의의。방법채용면역조직화학방법、단백인적법(Western blot)검측57례결직장암조직급거기암조직변연2 cm 이상적경하미견암침윤적정상조직중단백적표체정황。결과면역조직화학결과표명,CCNG2단백재결직장암조직중적표체교정상결직장조직명현강저,분별위38.6%(22/57)、91.3%(31/33)(P <0.05)。Western blot 결과표명,CCNG2적단백재결직장암조직적표체상대량교정상결직장조직적표체상대량명현강저(P <0.05)。CCNG2단백표체여결직장암 T 분기、림파결전이、종류림상분기급종류분화유관(P <0.05)。결론결직장암조직중 CCNG2단백표체명현감저,차여결직장암 T 분기、림파결전이、림상분기급종류분화유관。
Objective This study aimed to analyze the expression,clinical significance of cyclin G2(CCNG2)in colorectal carcinoma.Methods Immunohistochemistry and Western blot were used to analyze CCNG2 protein expression in 57 cases of colorectal cancer and normal tissues to explore the relationship between CCNG2 expression and clinical factors.Results Immunohistochemistry:the level of CCNG2 protein expression was significantly lower in colorectal cancer tissue than that of normal tissues,respectively 38.6% (22/57 )vs 91.3% (31/33 )(P < 0.05 ). Western blot:the relative amount of CCNG2 protein in colorectal cancer tissue was significantly lower than in normal tissues (P <0.05).The level of CCNG2 protein expression was correlated with T stages,lymph node metastasis,clinic stage and histological grade (P <0.05).Conclusion CCNG2 expression decreased in colorectal cancer and correlated significantly with colorectal cancer T stage,lymph node metastasis,clinic stage,histological grade,suggesting that CCNG2 may play important roles as a negative regulator to colorectal cancer.
