目的:观察四妙勇安汤合生脉饮对缺氧乳鼠心肌细胞活性及天冬氨酸氨基转移酶( AST)的影响。方法应用醇沉技术制备四妙勇安汤合生脉饮药液,采用胰蛋白酶和胶原酶联合消化的方法分离SD乳鼠心肌细胞,采取2次差速贴壁法对心肌细胞进行纯化,原代培养。利用高纯氮气建立心肌细胞缺氧/复氧模型。原代培养的乳鼠心肌细胞随机分为3组,即正常对照组、模型组及药物干预组。采用四甲基偶氮唑盐( MTT)比色法检测心肌细胞活性,采用全自动生化分析仪检测心肌细胞培养液AST的含量。结果药物干预组能提高乳鼠心肌细胞OD值,降低AST含量,与模型组比较差异有统计学意义( P<0.05)。结论四妙勇安汤合生脉饮可能提高缺氧/复氧损伤后心肌细胞的活性,降低AST含量,从而对心肌细胞具有保护作用。

목적:관찰사묘용안탕합생맥음대결양유서심기세포활성급천동안산안기전이매( AST)적영향。방법응용순침기술제비사묘용안탕합생맥음약액,채용이단백매화효원매연합소화적방법분리SD유서심기세포,채취2차차속첩벽법대심기세포진행순화,원대배양。이용고순담기건립심기세포결양/복양모형。원대배양적유서심기세포수궤분위3조,즉정상대조조、모형조급약물간예조。채용사갑기우담서염( MTT)비색법검측심기세포활성,채용전자동생화분석의검측심기세포배양액AST적함량。결과약물간예조능제고유서심기세포OD치,강저AST함량,여모형조비교차이유통계학의의( P<0.05)。결론사묘용안탕합생맥음가능제고결양/복양손상후심기세포적활성,강저AST함량,종이대심기세포구유보호작용。
Objective To observe the effect of Simiao Yongan decoction combined with Shengmai drink on AST and cardiomyocytes activity of hypoxic neonatal rat .Methods Simiao Yongan decoction with Shengmai drink was decocted and made by alcohol sedimentation .The combination of trypsin and collagenase was to isolate neonatal rat myocardial cells , which were purified twice by differential adhesion method and identified by immunocytochemical Methods Primary cultured neonatal rat cardiomyocytes were randomly divided into three groups , control group , model group , and treatment group .The myocardium viability was detected by MTT colorimetric method .And AST was determined by automatic biochemical analyzer .Results Drug intervention can improve the OD value of each well , lower AST levels, compared with the model group, the difference was significant (P<0.05).Conclusion The al-cohol sedimentation solution of Simiao Yongan decoction with Shengmai drink played a protective role on myocardial apoptosis, which may be relative with the improvement of the myocardium viability and the decline of the AST con-tent in culture media induced by hypoxia/reoxygenation treatment.
