【目的】探讨肥胖糖尿病(DM)患者脂肪细胞免疫受体核苷酸结合寡聚化结构域蛋白1(nucleotide-binding oligomerization domain-containing protein,NOD1)的表达与胰岛素抵抗的关系。【方法】取10例肥胖 DM患者(DM组)及10例非DM肥胖患者(NDM组)腹部皮下脂肪细胞,采用Real-time PCR方法检测两组脂肪细胞受体 NOD1的表达,加入 NOD1受体激动剂 iE-DAP,用酶联免疫吸附法(ELISA)检测两组细胞炎性细胞因子白细胞介素(IL)-6、IL-8及单核细胞趋化因子1(MCP-1)水平,2-脱氧-3 H-D-葡萄糖摄入法观察基础状态和 iE-DAP刺激状态的葡萄糖摄取率。【结果】DM组脂肪细胞 NOD1 mRNA的表达显著高于 NDM组(P<0.05);iE-DAP刺激下DM组 IL-6、IL-8及 MCP-1的分泌水平均显著高于 NDM组(P <0.05),DM组胰岛素刺激状态下脂肪细胞葡萄糖摄取率显著低于 NDM组(P<0.05)。【结论】NOD1介导的炎症反应可能参与肥胖糖尿病胰岛素抵抗的发生。

【목적】탐토비반당뇨병(DM)환자지방세포면역수체핵감산결합과취화결구역단백1(nucleotide-binding oligomerization domain-containing protein,NOD1)적표체여이도소저항적관계。【방법】취10례비반 DM환자(DM조)급10례비DM비반환자(NDM조)복부피하지방세포,채용Real-time PCR방법검측량조지방세포수체 NOD1적표체,가입 NOD1수체격동제 iE-DAP,용매련면역흡부법(ELISA)검측량조세포염성세포인자백세포개소(IL)-6、IL-8급단핵세포추화인자1(MCP-1)수평,2-탈양-3 H-D-포도당섭입법관찰기출상태화 iE-DAP자격상태적포도당섭취솔。【결과】DM조지방세포 NOD1 mRNA적표체현저고우 NDM조(P<0.05);iE-DAP자격하DM조 IL-6、IL-8급 MCP-1적분비수평균현저고우 NDM조(P <0.05),DM조이도소자격상태하지방세포포도당섭취솔현저저우 NDM조(P<0.05)。【결론】NOD1개도적염증반응가능삼여비반당뇨병이도소저항적발생。
[Obj ective]To explore the expression of immunologic receptor nucleotide-binding oligomerization domain-containing protein 1(NOD1)in adipocytes of obese patients with diabetes mellitus(DM)and its relation with insulin re-sistance(IR).[Methods]Abdominal subcutaneous adipocytes were drawn from 10 obese patients with DM(DM group) and 1 0 obese patients without DM(NDM group).Real-time PCR was used to detect the expression of NOD1 in adipocytes of two groups.NOD1 receptor agonist iE-DAP was administered.Enzyme linked immunosorbent assay(ELISA)was used to detect the levels of inflammatory cytokines such as interleukin-6(IL-6),interleukin-8(IL-8)and monocyte chemotactic factor-1(MCP-1)in two groups.The baseline state and iE-DAP-stimulated glucose uptake were observed by 2-deoxy-D-3 H glucose uptake assay.[Results]NOD1 mRNA expression in adipocytes of DM group was markedly higher than that of NDM group(P <0.05).The levels pf IL-6,IL-8 and MCP-1 in DM group with iE-DAP stimulation were markedly higher than those in NDM group(P<0.05).Adipocyte glucose transport rate in DM group with iE-DAP stim-ulation was significantly lower than that in NDM group(P<0.05).[Conclusion]NOD1-mediated inflammatory response may involve in the occurrence of IR in obese DM.
