目的:观察镇肝熄风汤对1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methy-4-phenyl-1,2,3,6-tetrahy-dropyridine ,MPTP)诱导的帕金森病( PD)模型小鼠脑黑质Bcl-2家族蛋白表达的影响。方法采用连续灌胃给予C57BL/6小鼠不同剂量镇肝熄风汤(12.5、25和50 g/kg)14天,于第11天开始,给药前1 h皮下注射MPTP(30 mg/kg),连续4天,造模后运用实时定量PCR法检测Bax、Bad、Bcl-2和Bcl-xl mRNA的表达。结果 PCR结果显示,镇肝熄风汤下调了Bax mRNA表达(P<0.05),同时上调了Bcl-2和Bcl-xl mRNA表达(P<0.05),对Bad mRNA表达无影响。结论镇肝熄风汤具有抑制PD模型小鼠脑黑质Bax mRNA表达,促进Bcl-2和Bcl-xL mRNA表达的作用。

목적:관찰진간식풍탕대1-갑기-4-분기-1,2,3,6-사경필정(1-methy-4-phenyl-1,2,3,6-tetrahy-dropyridine ,MPTP)유도적파금삼병( PD)모형소서뇌흑질Bcl-2가족단백표체적영향。방법채용련속관위급여C57BL/6소서불동제량진간식풍탕(12.5、25화50 g/kg)14천,우제11천개시,급약전1 h피하주사MPTP(30 mg/kg),련속4천,조모후운용실시정량PCR법검측Bax、Bad、Bcl-2화Bcl-xl mRNA적표체。결과 PCR결과현시,진간식풍탕하조료Bax mRNA표체(P<0.05),동시상조료Bcl-2화Bcl-xl mRNA표체(P<0.05),대Bad mRNA표체무영향。결론진간식풍탕구유억제PD모형소서뇌흑질Bax mRNA표체,촉진Bcl-2화Bcl-xL mRNA표체적작용。
Objective To investigate the effects of Zhenganxifeng decoction on expression of Bcl -2 family mRNA in the substantia nigra of MPTP-induced C57BL/6 mice.Methods C57BL/6 mice were given MPTP (30 mg/kg) to prepare Parkinson disease mice model .Different doses of Zhenganxifeng decoction (12.5, 25, and 50 g/kg) were given 14 d prior to MPTP in the pretreatment groups .Real-time PCR was used to analyze the expression of Bax , Bad, Bcl-2 and Bcl-xl mRNA.Results Zhenganxifeng decoction could down-regulate the expression of Bax mRNA , and up-regulate the expression of Bcl-2 and Bcl-xL mRNA.Conclusions The results suggest that Zhenganxifeng decoction could suppress the expression of Bax mRNA and Promote the expression of Bcl-2 and Bcl-xl mRNA in substantia nigra of Parkinson's disease mice induced by MPTP .
