目的:探讨巨噬细胞选择性活化与肥胖相关性胰岛素抵抗间的关系。方法28只3周龄雄性C57BL/6J小鼠,随机平均分为2组:普食组(A组,n=14只),高脂饮食组(B组,n=14只),分别给予标准饲料与高脂饲料喂养。6周、12周分别检测空腹血糖((FBG)、血浆胰岛素(FIns)水平,并计算胰岛素抵抗指数(HOMA-IR),测定肝组织匀浆三酰甘油(TG)、胆固醇(TC)及丙氨酸氨基转移酶(ALT)水平;HE染色观察肝脏及脂肪组织病理学改变,DAB染色及免疫荧光染色检测小鼠肝脏、脂肪组织M2型巨噬细胞的特异性蛋白Ym-1表达情况。结果第6周B组小鼠体质量、FIns、HOMA-IR、TC均明显高于A组,FBG、TG、ALT较A组升高不明显;第12周B组小鼠体质量、FBG、FIns、HOMA-IR、TC、TG和ALT进一步升高,均明显高于A组。第12周B组小鼠脂肪组织内巨噬细胞选择性活化标志Ym-1蛋白表达较A组增强。结论巨噬细胞选择性活化可能参与了肥胖诱导胰岛素抵抗的发生。

목적:탐토거서세포선택성활화여비반상관성이도소저항간적관계。방법28지3주령웅성C57BL/6J소서,수궤평균분위2조:보식조(A조,n=14지),고지음식조(B조,n=14지),분별급여표준사료여고지사료위양。6주、12주분별검측공복혈당((FBG)、혈장이도소(FIns)수평,병계산이도소저항지수(HOMA-IR),측정간조직균장삼선감유(TG)、담고순(TC)급병안산안기전이매(ALT)수평;HE염색관찰간장급지방조직병이학개변,DAB염색급면역형광염색검측소서간장、지방조직M2형거서세포적특이성단백Ym-1표체정황。결과제6주B조소서체질량、FIns、HOMA-IR、TC균명현고우A조,FBG、TG、ALT교A조승고불명현;제12주B조소서체질량、FBG、FIns、HOMA-IR、TC、TG화ALT진일보승고,균명현고우A조。제12주B조소서지방조직내거서세포선택성활화표지Ym-1단백표체교A조증강。결론거서세포선택성활화가능삼여료비반유도이도소저항적발생。
Objective To explore the relationship between the selective activation of macrophages and insu -lin resistance.Methods 28 male C57BL/6J mice were randomly assigned into 2 groups.the standard chow group (group A,n=14),mice were fed with basic diet,and high-fat diet-fed group (group B,n=14),mice were t fed with high-fat diet.At 6 and 12 weeks,the levels of fasting peripheral blood glucose (FBG),fasting plasma insulin (FIns) were detected and the levels of triglyceride ( TG) ,cholesterol ( TC) and glutamic pyruvic transaminase ( ALT) were also measured.The changes in adipose tissue and liver were observed .The expression of Ym-1proteinin was detected . Results Compared with group A,the body mass,the level of FIns,HOMA-IR,and TC in group B were significantly increased at the 6th week.However,there were no statistically differences in the levels of FBG ,TG and ALT.The body mass,the levels of FBG,FIns,HOMA-IR,TC,TG and ALT in group B were significantly higher than those in the group A at the 12th week.The expression of macrophages specific marker Ym-1 in adipose tissue was significantly in-creased in group B at the 12th week compared with that in group A .Conclusion Selective activation of macrophage may play a role in the development of obesity-related insulin resistance .
