目的:快速建立大鼠动脉粥样硬化( Arteriosclerosis,AS)模型,并观察茶多酚对AS大鼠的防治作用及机制.方法将60只Wistar大鼠随机分为正常对照组(NC组)、模型组(AS组)、维生素E组(VE组,0.2 g/kg)、茶多酚高剂量组(TP1组,0.4 g/kg)、茶多酚低剂量组(TP2组,0.2 g/kg),干预9周,测定大鼠血脂水平及抗氧化能力.结果1)AS组与正常对照组相比,各项检测指标之间比较均具有统计学意义(P<0.05);2)血脂:维生素E组、茶多酚高剂量组、茶多酚低剂量组与AS组相比,血清总胆固醇( TC)、甘油三酯( TG)、低密度脂蛋白胆固醇(LDL-C)含量均降低(P<0.05),高密度脂蛋白(HDL-C)含量显著增高(P<0.05);3)血清超氧化物歧化酶( SOD)、丙二醛( MDA)、一氧化氮合酶( NOS)水平:维生素E组、茶多酚高剂量组、茶多酚低剂量组与AS组比较,血清SOD,NOS活力升高(P<0.05),MDA含量降低(P<0.05).结论茶多酚对大鼠具有防治AS作用,其作用机制与降低血脂、清除氧自由基、增强NOS活性、调节NO的代谢、保护血管内皮细胞有关.

목적:쾌속건립대서동맥죽양경화( Arteriosclerosis,AS)모형,병관찰다다분대AS대서적방치작용급궤제.방법장60지Wistar대서수궤분위정상대조조(NC조)、모형조(AS조)、유생소E조(VE조,0.2 g/kg)、다다분고제량조(TP1조,0.4 g/kg)、다다분저제량조(TP2조,0.2 g/kg),간예9주,측정대서혈지수평급항양화능력.결과1)AS조여정상대조조상비,각항검측지표지간비교균구유통계학의의(P<0.05);2)혈지:유생소E조、다다분고제량조、다다분저제량조여AS조상비,혈청총담고순( TC)、감유삼지( TG)、저밀도지단백담고순(LDL-C)함량균강저(P<0.05),고밀도지단백(HDL-C)함량현저증고(P<0.05);3)혈청초양화물기화매( SOD)、병이철( MDA)、일양화담합매( NOS)수평:유생소E조、다다분고제량조、다다분저제량조여AS조비교,혈청SOD,NOS활력승고(P<0.05),MDA함량강저(P<0.05).결론다다분대대서구유방치AS작용,기작용궤제여강저혈지、청제양자유기、증강NOS활성、조절NO적대사、보호혈관내피세포유관.
Objective To rapidly develop rat ’ s arteriosclerosis model ( AS ) for observing the effect of tea polyphenol on rats with AS and its mechanism. Methods Sixty Wistar rats were randomly divided into normal control group(NC group),AS group,vitamin E group(VE group,0. 2g/kg),tea polyphenol high-dose group(TP1 group,0. 4g/kg),and tea polyphenol low-dose group(TP2 group,0. 2g/kg). At the end of the 9th week,the level of blood lipid and the anti-oxidant ability were examined. Results ( 1 ) Compared of AS group and normal control group,each testing index in AS group were significantly different(P<0. 05);(2) Blood lipid:compared with AS group,serum contents of TC,TG and LDL-C in vitamin E group,tea polyphenol high-dose group,and tea polyphenol low-dose group were lower(P<0. 05),and HDL-C were higher(P<0. 05);(3) Serum levels of SOD, MDA,NOS:vitamin E group,tea polyphenol high-dose group and tea polyphenol low-dose group compared with AS group,the activities of SOD and NOS were higher(P<0. 05),and the content of MDA was lower(P<0. 05). Conclusion Tea polyphenol exhibits preventive and therapeutic effects on rat with AS,and the mechanisms may be related to its lowering blood lipid, clearing oxygen free radicals, enhancing NOS activity, regulating NO metabolism and protecting vascular endothelium from injury.
