考察了海藻酸钠法制备缬氨酸转氨酶固定化细胞的制备条件,以及该固定化细胞的操作稳定性。正交试验结果显示:固定化细胞制备条件为海藻酸钠浓度30 g/L、交联剂戊二醛浓度1.5%(V/V)、菌体负载量40 g/L、固定化时间4 h。该固定化细胞连续使用10次,底物平均转化率为15.48%,显示了较好的稳定性。
攷察瞭海藻痠鈉法製備纈氨痠轉氨酶固定化細胞的製備條件,以及該固定化細胞的操作穩定性。正交試驗結果顯示:固定化細胞製備條件為海藻痠鈉濃度30 g/L、交聯劑戊二醛濃度1.5%(V/V)、菌體負載量40 g/L、固定化時間4 h。該固定化細胞連續使用10次,底物平均轉化率為15.48%,顯示瞭較好的穩定性。
고찰료해조산납법제비힐안산전안매고정화세포적제비조건,이급해고정화세포적조작은정성。정교시험결과현시:고정화세포제비조건위해조산납농도30 g/L、교련제무이철농도1.5%(V/V)、균체부재량40 g/L、고정화시간4 h。해고정화세포련속사용10차,저물평균전화솔위15.48%,현시료교호적은정성。
The preparation conditions of immobilized cells with valine-pyruvate transaminase activity are studied.The optimal temperature,pH and operational stability of immobilized cells are also inves-tigated.The results show that the optimal conditions for preparation of immobilized cells are sodium alginate of 30 g/L,glutaraldehyde of 1.5% (V/V),cells loaded concentration of 40 g/L,cells immo-bilization time of 4 h.The immobilized cells are continuously used for 10 times and the average con-version rate is 15.48%,which shows better stability.
