目的:探讨组蛋白 H3第9位赖氨酸残基乙酰化(H3K9ac)及组蛋白 H3第4位赖氨酸残基二甲基化(H3K4me2)在卵巢浆液性囊腺瘤、卵巢交界性浆液性囊腺瘤和卵巢浆液性囊腺癌组织中蛋白表达与浆液性上皮性卵巢癌发生、发展的关系及两者之间有无相关性。方法:应用免疫组织化学链霉菌抗生物素蛋白-过氧化酶连接法(SP 法)检测 H3K9ac、H3K4me2在30例卵巢浆液性囊腺瘤组织、30例卵巢交界性浆液性囊腺瘤组织及40例卵巢浆液性囊腺癌组织中的表达,观察表达差异并进行统计学处理。结果:H3K9ac 在卵巢浆液性囊腺瘤组织中阳性表达率为93.33%,在卵巢交界性浆液性囊腺瘤组织中阳性表达率为66.67%,在卵巢浆液性囊腺癌中阳性表达率为42.50%,良性、交界性、恶性组织间两两比较差异有统计学意义( P ﹤0.05)。在不同手术病理分期、不同组织学分级的卵巢浆液性囊腺癌组织中 H3K9ac 的阳性表达率有统计学差异(P ﹤0.05)。H3K4me2在卵巢浆液性囊腺瘤组织中阳性表达率为90%,在交界性浆液性囊腺瘤组织中阳性表达率为73.33%,在卵巢浆液性囊腺癌中阳性表达率为52.50%,良性与交界性、交界性与恶性组织间比较差异无统计学意义(P ﹥0.05),良性与恶性组织间比较差异有统计学意义( P ﹤0.05)。在不同手术病理分期、不同组织学分级的卵巢浆液性囊腺癌组织中 H3K4me2的阳性表达率无统计学差异( P ﹥0.05)。H3K9ac、H3K4me2在卵巢浆液性囊腺癌组织中的表达呈正相关(r =0.732,P ﹤0.001)。结论:H3K9ac、H3K4me2的低表达与浆液性上皮性卵巢癌的发生、发展有关,有可能成为浆液性上皮性卵巢癌诊断和治疗的新靶点。

목적:탐토조단백 H3제9위뢰안산잔기을선화(H3K9ac)급조단백 H3제4위뢰안산잔기이갑기화(H3K4me2)재란소장액성낭선류、란소교계성장액성낭선류화란소장액성낭선암조직중단백표체여장액성상피성란소암발생、발전적관계급량자지간유무상관성。방법:응용면역조직화학련매균항생물소단백-과양화매련접법(SP 법)검측 H3K9ac、H3K4me2재30례란소장액성낭선류조직、30례란소교계성장액성낭선류조직급40례란소장액성낭선암조직중적표체,관찰표체차이병진행통계학처리。결과:H3K9ac 재란소장액성낭선류조직중양성표체솔위93.33%,재란소교계성장액성낭선류조직중양성표체솔위66.67%,재란소장액성낭선암중양성표체솔위42.50%,량성、교계성、악성조직간량량비교차이유통계학의의( P ﹤0.05)。재불동수술병리분기、불동조직학분급적란소장액성낭선암조직중 H3K9ac 적양성표체솔유통계학차이(P ﹤0.05)。H3K4me2재란소장액성낭선류조직중양성표체솔위90%,재교계성장액성낭선류조직중양성표체솔위73.33%,재란소장액성낭선암중양성표체솔위52.50%,량성여교계성、교계성여악성조직간비교차이무통계학의의(P ﹥0.05),량성여악성조직간비교차이유통계학의의( P ﹤0.05)。재불동수술병리분기、불동조직학분급적란소장액성낭선암조직중 H3K4me2적양성표체솔무통계학차이( P ﹥0.05)。H3K9ac、H3K4me2재란소장액성낭선암조직중적표체정정상관(r =0.732,P ﹤0.001)。결론:H3K9ac、H3K4me2적저표체여장액성상피성란소암적발생、발전유관,유가능성위장액성상피성란소암진단화치료적신파점。
To explore the protein expression of H3K9ac and H3K4me2 in ovarian serous cystadenoma tissues,borderline serous cystadenoma tissues and serous cystadenocarcinoma tissues,their roles in occurrence and de-velopment of ovarian serous tumors,and the relationship between H3K9ac and H3K4me2. Methods:The expressions of H3K9ac,H3K4me2 in 30 cases of ovarian serous cystadenoma tissues,30 cases of ovarian borderline serous cystad-enoma tissues,40 cases of ovarian serous cystadenocarcinoma tissues were detected by immunology and histology chemistry SP. Results:There were significant differences in the expression of H3K9ac in ovarian serous cystadenoma tissues,ovarian borderline serous cystadenoma tissues,and ovarian serous cystadenocarcinoma tissues,and had correl-ativity with surgical pathologic stage and histological grade(P ﹤ 0. 05). The expression of H3K4me2 was significantly lower in ovarian serous cystadenocarcinoma than that in ovarian serous cystadenoma(P ﹤ 0. 05),but had no correlativ-ity with surgical pathologic stage and histological grade(P ﹥ 0. 05). H3K9ac had positive correlation with H3K4me2 in ovarian serous cystadenocarcinoma(r = 0. 732,P ﹤ 0. 001). Conclusion:H3K9ac and H3K4me2 protein expression is decreased in ovarian serous cystadenocarcinoma. The expression of H3K9ac and H3K4me2 might be correlated to the carcinogenesis and development of ovarian epithelial cancer. They might become the new target for diagnosis and treatment of ovarian epithelial cancer.
