本试验旨在用聚合酶链式反应-变性梯度凝胶电泳( PCR-DGGE)技术分析不同生长性能生长猪的肠道菌群差异。试验以36头体重为(26.6±2.6)kg的“杜×长×大”阉公猪为材料,进行42 d单栏饲养(自由采食),每周记录采食量与体重并计算饲料转化率以及平均日增重。试验结束时综合考虑饲料转化率以及平均日增重将36头猪分为3组:高、中、低生长性能组,每组选择3头进行屠宰(共9头),分别收集回肠、盲肠、结肠食糜以及粪样,通过PCR-DGGE技术测定菌群组成。结果表明:与生长性能低的猪相比,生长性能高的猪回肠、结肠、盲肠和粪中细菌的DGGE条带数增加;盲肠中粪球菌、结肠和粪样中罗氏菌属细菌的丰度均升高,并且在个别生长性能高的猪盲肠中罗氏菌属细菌和乳酸菌为优势细菌。因此,猪的肠道微生物组成与猪的生长性能有一定关联;粪球菌、罗氏菌属细菌和乳酸菌在生长性能高的猪肠道中是优势菌。
本試驗旨在用聚閤酶鏈式反應-變性梯度凝膠電泳( PCR-DGGE)技術分析不同生長性能生長豬的腸道菌群差異。試驗以36頭體重為(26.6±2.6)kg的“杜×長×大”閹公豬為材料,進行42 d單欄飼養(自由採食),每週記錄採食量與體重併計算飼料轉化率以及平均日增重。試驗結束時綜閤攷慮飼料轉化率以及平均日增重將36頭豬分為3組:高、中、低生長性能組,每組選擇3頭進行屠宰(共9頭),分彆收集迴腸、盲腸、結腸食糜以及糞樣,通過PCR-DGGE技術測定菌群組成。結果錶明:與生長性能低的豬相比,生長性能高的豬迴腸、結腸、盲腸和糞中細菌的DGGE條帶數增加;盲腸中糞毬菌、結腸和糞樣中囉氏菌屬細菌的豐度均升高,併且在箇彆生長性能高的豬盲腸中囉氏菌屬細菌和乳痠菌為優勢細菌。因此,豬的腸道微生物組成與豬的生長性能有一定關聯;糞毬菌、囉氏菌屬細菌和乳痠菌在生長性能高的豬腸道中是優勢菌。
본시험지재용취합매련식반응-변성제도응효전영( PCR-DGGE)기술분석불동생장성능생장저적장도균군차이。시험이36두체중위(26.6±2.6)kg적“두×장×대”엄공저위재료,진행42 d단란사양(자유채식),매주기록채식량여체중병계산사료전화솔이급평균일증중。시험결속시종합고필사료전화솔이급평균일증중장36두저분위3조:고、중、저생장성능조,매조선택3두진행도재(공9두),분별수집회장、맹장、결장식미이급분양,통과PCR-DGGE기술측정균군조성。결과표명:여생장성능저적저상비,생장성능고적저회장、결장、맹장화분중세균적DGGE조대수증가;맹장중분구균、결장화분양중라씨균속세균적봉도균승고,병차재개별생장성능고적저맹장중라씨균속세균화유산균위우세세균。인차,저적장도미생물조성여저적생장성능유일정관련;분구균、라씨균속세균화유산균재생장성능고적저장도중시우세균。
This experiment was conducted to analysis the differences of intestinal microbial composition in pigs with different growth performances by polymerase chain reaction-denaturing gradient gel electrophoresis( PCR-DGGE). Thirty-six barrows(Duroc×Landrace×Yorkshire)with a starting body weight of(26.6±2.6)kg were raised for 42 days( ad libitum)within single bar. Feed intake and body weight of each pig were recorded every week to calculate feed conversion rate and average daily gain. At the end of the experiment,feed conver-sion ratio and average daily gain were integrated to divide the pigs into 3 groups,namely,high growth per-formance group,middle growth performance group and low growth performance group. Three selected pigs from each group were killed to collect gut contents and feces. The results showed that the DGGE band numbers of ileum,colon,cecum and fecal samples were increased with the improvement of growth performance. Spe-cifically,the abundance of Coprococcus sp. in cecum,Roseburia sp. in colon and fecal samples were higher in high growth performance group than those in the low growth performance group,while Roseburia sp. and Lac-tobacillus sp. were abundant in the gut of some high growth performance pigs. In summary,there is a correla-tion between pig gut microbiota composition and the growth performance of pigs. Bacteria belong to the genus of Coprococcus,Roseburia,Lactobacillus can serve as target for improving the health and growth performance of pigs.
