VEGF-C%Non-small cell lung cancer
目的:探讨沉默血管内皮生长因子( VEGF-C)基因对人非小细胞肺癌( NSCLC) A549细胞增殖和侵袭能力的影响。方法构建靶向VEGF-C基因的重组慢病毒Lv-siRNA-VEGF-C,将其感染A549细胞,建立VEGF-C基因低表达的A549细胞。 Western印迹检测Lv-siRNA-VEGF-C细胞组、空载体Lv-Ctrl 对照组及空白A549细胞中VEGF-C蛋白的表达噻唑蓝( MTT)法、Transwell侵袭实验和裸鼠移植瘤模型分别检测各组细胞的增殖和迁移能力。结果与对照相比,VEGF-C基因低表达组细胞中VEGF-C蛋白的表达显著降低,细胞体内、体外增殖活性也明显下降,细胞侵袭能力亦显著降低。结论成功构建的慢病毒介导的针对VEGF-C的干扰载体可有效抑制A549细胞表达 VEGF-C 蛋白表达,沉默 VEGF-C能抑制人NSCLC A549细胞的增殖和侵袭能力。

Objective To explore the effect of silencing vascular endothelial growth factor ( VEGF)-C gene expression on prolifera-tion and invasion of non-small cell lung cancer (NSCLC) A549 cell.Methods The recombinant lentivirus Lv-siRNA-VEGF-C targeting VEGF-C gene was constructed and then was transfected into A 549 cells to establish VEGF-C low expressed cells .The expression level of VEGF-C protein in A549 cells was examined by Western blot in Lv-siRNA-VEGF-C, Lv-Con control and blank control groups .The prolifera-tion and invasion of A549 cells were detected by MTT, transwell analysis and human lung cancer xenograft respectively .Results Compared with that of control group ,the expression of VEGF-C protein in A549 cells was significantly lower in Lv-siRNA-VEGF-C A549 cells, and the proliferation activity in vivo and in vitro of low VEGF-C expressed cells were also decreased significantly .Simutanously,the invasion of low expressed VEGF-C A549 cells was significantly increased .Conclusions Lentvirus-mediated silencing VEGF-C gene could inhibit the prolif-eration, migration and invasion of human non-small cell lung cancer A549 cells.
