目的:目的研究BDNF对大鼠急性脊髓损伤后Bcl-2表达变化的影响。方法参照Nystrom压迫方法制作大鼠脊髓压迫损伤模型,成年健康Wistar大鼠88只,雌雄不限,随机分为正常对照组8只、损伤组40只、BDNF组80只。免疫组化和 Western 印迹检测各组大鼠 Bcl-2的表达。结果免疫组化结果显示:正常对照组Bcl-2阳性产物的平均光密度值(MOD)显著高于损伤组和BDNF组(P<0.05);而损伤组与BDNF组相比MOD值明显降低(P<0.05);Western 印迹结果显示:与正常对照组比较,损伤组与BDNF组Bcl-2的灰度值( IDV)与内参照IDV的比值明显降低(P<0.05),而损伤组则明显低于BDNF组(P<0.05)。结论 BDNF很可通过上调Bcl-2的表达参与脊髓继发性损伤。

Objective To study the expression of Bcl-2 in acute spinal cord injury rat after BDNF injected .Methods The spinal cord injury was induced by Nystrom's way.The healthy adult Wistar rats (88) were randomly divided into normal control , spinal cord injury, BDNF groups on average .The expression of Bcl-2 was observed by immunochemistry and Western blot .Results The mean optic density ( MOD) of Bcl-2 positive product was decreased significantly in acute spinal cord injury group and BDNF group than that in normal control (P<0.05);but increased in BDNF group than that in acute spinal cord injury group (P<0.05).The integrated density value (IDV) was decreased significantly in spinal cord injury and BDNF groups than that in normal control group (P<0.05);but increased in BDNF group than that in spinal cord injury group (P<0.05).Conclusions One of the mechanisms for BDNF participate the pathogenesis of acute spinal cord injury mightbe upregulate the expression of Bcl-2.
