目的:探讨归肾丸对卵巢储备功能低下(diminished ovarian reserve ,DOR)小鼠的颗粒细胞超微结构的影响。方法将20只C57 BL/6J雌性小鼠随机分为正常组、模型组、归肾丸组和脱氢表雄酮(DHEA)组。连续序贯腹腔注射孕马血清(PMSG)、人绒毛膜促性腺激素(HCG)、前列腺素 F2α(PGF2α)产生连续超数排卵,再结合臭氧吸入构建DOR模型。其后各治疗组给予相应药物灌胃,而正常组与模型组给予生理盐水灌胃。21 d后,采用透射电镜观察各组小鼠卵巢颗粒细胞超微结构的变化。结果模型组中线粒体肿胀明显,还出现了空泡化改变,且较少看到脂滴,各治疗组卵巢超微结构较模型组有明显差异,颗粒细胞线粒体形态规则,管型明显,胞质内脂滴丰富。结论归肾丸对 DOR模型小鼠卵巢超微结构有较明显的改善作用。

목적:탐토귀신환대란소저비공능저하(diminished ovarian reserve ,DOR)소서적과립세포초미결구적영향。방법장20지C57 BL/6J자성소서수궤분위정상조、모형조、귀신환조화탈경표웅동(DHEA)조。련속서관복강주사잉마혈청(PMSG)、인융모막촉성선격소(HCG)、전렬선소 F2α(PGF2α)산생련속초수배란,재결합취양흡입구건DOR모형。기후각치료조급여상응약물관위,이정상조여모형조급여생리염수관위。21 d후,채용투사전경관찰각조소서란소과립세포초미결구적변화。결과모형조중선립체종창명현,환출현료공포화개변,차교소간도지적,각치료조란소초미결구교모형조유명현차이,과립세포선립체형태규칙,관형명현,포질내지적봉부。결론귀신환대 DOR모형소서란소초미결구유교명현적개선작용。
Objective To investigate the effects of Guishenwan on ovarian ultrastructure in mice with dimin-ished ovarian reserve(DOR).Methods 16 female C57 BL/6J mice were randomly divided into 4 groups ,including normal group ,model group ,Guishenwan group and DHEA group.Sequentially administrating pregnant mare serum gonadotropin(PMSG) ,human chorionic gonadotropin(HCG) ,and prostaglandin F2α(PGF2α)and exposing to ozone were used to induce DOR model.Mice in Guishenwan group and DHEA group were intragastrically administered with corresponding drugs while mice in normal group and model group received the same amount of saline for 21 days.T he ultrastructure of ovary w as observed by transmission electron microscope.Results T he ovarian ultra-structure of rats in model group was obviously different with those in Guishenwan group and DHEA group.In granule cells of model group ,the mitochondria was swell ,vacuolation ,and lipid drop was decreased ,while granule cells of Guishenwan group and DHEA group had more regular mitochondrial morphology and were richer in lipid drops.Conclusion Guishenwan has an obvious improvement on ovarian ultrastructure in mice with DOR.
